摘  要:目的建立高效液相色谱法(HPLC)同时测定正柴胡饮颗粒中升麻素苷和5-O-甲基维斯阿米醇苷含量的方法。方法色谱柱为Agilent C18(150mm×4.6mm,5μm),柱温30℃;流动相为乙腈-0.05%磷酸(16∶84),流速为1.0ml/min;检测波长254nm。结果升麻素苷和5-O-甲基维斯阿米醇苷的回归方程分别为Y=28.8653X+22.8925(r=0.9994)和Y=39.8631X+9.3742(r=0.9996);线性范围分别为13.95-139.5μg/ml和13.02-130.2μg/ml;平均回收率分别为98.11%(RSD=1.06%)和98.05%(RSD=0.66%)。结论该HPLC测定操作简便,结果精确,重现性好,可用于正柴胡饮颗粒的质量评价。Objective To establish a high performance liquid chromatography(HPLC)method for simultaneous determination of the contents of prim-O-glucosylcimifugin and 5-O-methylvisamminoside in Zhengchaihuyin particals.Methods Agilent C18column(150mm×4.6mm,5μm)was used at 30℃.The mobile phase consisted of a mixture of acetonitrile-0.05% phosphoric acid solution(16∶84)in a flow rate of 1.0ml/min.The detection wave length was set at 254nm.Results The regression equations for prim-O-glucosylcimifugin and 5-O-methylvisamminoside were Y=28.8653X +22.8925(r=0.9994)and Y=39.8631X+9.3742(r=0.9996),with the linear ranges of 13.95139.5μg/ml(r=0.9994)and 13.02-130.2μg/ml(r=0.9996),respectively.Moreover,the mean recovery rates of the two constituents were 98.11%(RSD=1.06%)and 98.05%(RSD=0.66%),respectively.Conclusion The method mentioned above is simple and reproducible with excellent accuracy and reliability for determination of prim-O-glucosylcimifugin and 5-O-methylvisamminoside and can be used for evaluating the quality of Zhengchaihuyin particals.
