摘  要:目的观察丹参多酚酸盐对人肾小管上皮细胞血管性血友病因子裂解酶表达的影响,初步分析其意义。方法肾小管上皮细胞体外培养,将细胞接种在培养瓶中,待细胞融合至80%~90%时,更换含有丹参多酚酸盐终浓度分别为0、0.1、0.5、1.0、2.0mg/L的培养基,培养24h后,收集人肾小管上皮细胞并提取mRNA,转化为CDNA,用实时定量荧光聚合酶链反应技术(RT-PCR)检测人肾小管上皮细胞血管性血友病因子裂解酶mRNA的表达,并经凝胶成像行半定量分析。结果以空白对照人肾小管上皮细胞表达的血管性血友病因子裂解酶mRNA量为参照1,则0.1、0.5、1.0、2.0mg/L浓度丹参多酚酸盐共培养后的人肾小管上皮细胞表达的血管性血友病因子裂解酶mRNA依次为2.39、3.05、4.65和6.65,数据为重复2次实验的平均值。结论人肾小管上皮细胞可表达血管性血友病因子裂解酶mRNA,丹参多酚酸盐能增加人肾小管上皮细胞血管性血友病因子裂解酶的表达。丹参多酚酸盐对肾小管上皮细胞血管性血友病因子裂解酶的调节可能会影响肾脏局部循环的出血、凝血状态,可能是丹参多酚酸盐的药理机制之一。Objective This study was to observe the effect of salvianolate on the expression of von Willebrand factor cleaving protease (vWF-CP) in human renal proximal tubular epithelial cells (human kidney-2, HK2) for preliminarily analyzing the significance. Methods HK2 cells were culti- vated in culture flasks. When the conflueney of the cells was 80%-90%, we replaced with the medi- um containing salvianolate contents of 0, 0. 1, 0. 5, 1.0 and 2. 0 mg/L, respectively. After culturing for 24 hours, the collected cells were extracted for mRNA, which was then converted to cDNA. The mRNA levels of vWFCP of HK2 were detected by RT-PCR. Gel images were analyzed semi-quantita- tively. Results Taken the HK2 with no salvianolate co-cultured as control, the mRNA levels of vWF-CP of HK2 co-cultured by the medium containing salvianolate contents of 0. 1, 0. 5, 1.0 and 2. 0 mg/L were 2. 39, 3. 05, 4. 65 and 6. 65 respectively, which were the average of two repetitive tests. Conclusions In mRNA level, HK2 could express vWF-CP, while salvianolate could increase the vWF-CP expression in a dose-dependent manner. The regulation of salvianolate to vWF-CP expressed in renal tubular epithelial cells may affect the coagulation of kidney local circulation, which could be one of pharmaco-mechanisms of salvianolate.
