摘 要:将瑶山苣苔无菌苗幼叶接种于添加不同植物生长调节剂(PGRs)和活性炭(AC)的MS培养基中,研究PGRs和AC对叶片直接诱导丛生芽及幼芽生根的影响,并建立瑶山苣苔的植株再生体系。结果表明:叶片外植体在MS+1.0mg·L-16-BA+0.1mg·L-1NAA培养基上的丛生芽诱导率较高(96.43%);MS培养基中添加0.1%AC时,丛生芽的诱导率为83.67%,但丛生芽比添加PGRs诱导出来的生长健壮;在1/2MS+0.1mg·L-1NAA+2%蔗糖的培养基上,不定根诱导率高达100%;1/2MS培养基中添加AC,幼苗的生长比在添加NAA的培养基上的健康;幼苗经炼苗后移栽到混合基质(沙子:蛭石:珍珠岩=1:1:1)中成活率可达85%。The objective of this study was to investigate the direct effect of different culture media with a series of concentrations of plant growth regulators (PGRs) and activated charcoal (AC) on initiation of multiple shoots and budlet rooting, then the regeneration system ofDayaoshania cotinifolia was established. The explants used in this study were the young leaves of aseptic seedlings obtained from the seeds of D. cotinifolia. The results showed that the explants inoculated on MS medium with 1.0 mg.L-1 6-BA and 0.1 mg-L-1 NAA were provided with a relatively high induction rate about 96.43% of the multiple shoots. The MS medium added with 0.1% AC was also found to be beneficial to the growth of multiple shoots. Meanwhile, the buds inoculated on 1/2MS+0.1 mg.L-1 NAA+2% sucrose can also obtain a 100% ideal induction rate of adventitious roots. Moreover, the seedlings grew better than the ones grew on the 1/2MS medium with NAA when AC was added in the medium with a proper concentration. As much as 85% of plantlets survived following acclimatization and transfer to potting mixture (sand:vermiculite:perlite= 1:1:1).
