纤维蛋白胶携载的粒细胞集落刺激因子促进大鼠缺血后肢血管再生

摘　　要：目的观察纤维蛋白胶携载的粒细胞集落刺激因子（Gel＋G-CSF）对大鼠外周血白细胞总数和缺血下肢血管再生的影响。方法雄性sD大鼠30只，制成右下肢缺血模型，随机分成3组：Gel＋G-CSF组、G-CSF组和PBS组，每组10只，分别肌肉注射Gel＋G-CSF、G-CSF和PBS。造模术前及术后第1、3、5、7天计数外周血白细胞总数，造模后第1周和第4周每组分别处死5只大鼠，行组织学检测，计数血管密度。结果Gel＋G-CSF组外周血白细胞总数在术后第1天达峰值[（14．694±1．11）×10^9／L]，且明显低于G．CSF组在术后第3天达到的峰值[（21．00±2．26）×10^9／L]。Gel＋G-CSF组[（686±108）／mm^2]的毛细血管密度明显高于G-CSF组[（491±110）／mm^2]和PBS组[（252±78）／mm^2]，P〈0．05。Gel＋G-CSF组[（6．14-0．8）／mm^2]的α-SMA阳性血管密度明显高于PBS组[（2．6±1．3）／mm^2]，P〈0．05。Gel＋G-CSF组肌组织中有大量血管内皮生长因子阳性细胞的浸润。结论Gel＋G-CSF可以促进缺血肌组织血管再生，而刺激外周血白细胞计数升高的不良反应较小。Objective To observe the effect of granulocyte colony-stimulating factor carried byfibrin gel on the total number of peripheral white blood cells and neovascularization of ischemic muscle in rat hindlimb ischemia model. Methods Thirty male SD rats were subjected to right hindlimb ischemia and randomly divided into three groups： Gel ＋ G-CSF, G-CSF, and PBS, respectively injected with Gel ＋ G-CSF, G-CSF and PBS. WBCs was detected before and 1,3,5,7 days; At first week and fourth week after surgery, 5 rats in each group were sacrificed, and after histological detections was performed, blood vessels density counted. Results The peak value of WBCs counts in group Gel ＋ G-CSF （ 14. 69 ± 1.11×10^9/L） appeared at postoperative day 1 was significantly lower than that in group G-CSF （21.00 ± 2.26×10^9/L） at postoperative day 3. The capillary density in group Gel ＋ G-CSF （ 686 ± l0^8/mm2 ） was significantly higher than that in group G-CSF （491± ll0/mm^2） and group PBS （252±78/mm2）, P 〈0.05. The ct-SMA- positive blood vessel density in group Gel ＋ G-CSF （ 6. 1± 0. 8/mm^2 ） was significantly higher than that in group PBS （ 2. 6 ±1.3/mm^ 2）, P 〈 O. 05. In group Gel ＋ G-CSF, there were many VEGF-positive cells infiltrating in ischemic limb. Conclusions Gel ＋ G-CSF promotes neovaseularization in isehemic muscle, and induced more modest WBCs counts increase than the treatment with G-CSF.
