摘 要:富血小板血浆(plateletrichplasma,PRP)含多种生长因子,如转化生长因子β(transforminggrowthfactor,TGF-β)、血管内皮生长因子、血小板源性生长因子、表皮生长因子、胰岛素样生长因子等。国内外大量文献[3-4]报道PRP可促进骨缺损的修复,并在体外对骨髓基质干细胞(bone marrowstromal stem cells,BMSCs)的增殖与分化有明显促进作用,但也有少数学者№圳通过动物实验认为PRP无促进骨再生作用,在临床应用中PRP复合植骨材料的修复效果也不明显。Objective To investigate whether autologous platelet-ric.h plasma (PRP) carl be re- pla~'ed hy allogeneic PRP to promote the in vitro proliferation and differentiation of bone marrow stromal stem cells (BMSCs) in rabbits and to find an optimal concentration at which allogeneic PRP can best promote the prolil^ration and differentiation of BMSCs. Methods Three New Zealand rabbits were used for conven- lional cuhure of BMSCs and preparation of PRP. Experiments were conducted in 4 groups with 4 kinds of ctdture medium: hlauk group (DMEM only), conventional group I I)MEM containing 10% fetal bovine serum (FBS) ], autologous group (DMEM containing 10% FBS plus autologous PRP extract) and allngeneic group (I)MEM containing 10% FBS plus allogeneic PRP extract). The allogeneic group was further divided into five suhgrottps according to the cmlcentratiuns of PRP extract in the medium: 10%, 20%, 30%, 40% and 50% respectively. After the best concentration was determined in the allogeneic group, it was used in the autolo- gous group to compare the effects of the 2 kinds of PRP. On clay 4, 8, 12, 16 and 20 after intervention, optical density (OD) value in M3T assay and alkaline phosphatase (ALP) activity were tested in all groups and the cell apoptosis rate at the best concentration of PRP extract was a]so assessed in each group. Results The effect of PRP on the proliferation and differenliatiou of BMSCs was negatively related to the eoncen- lraliou of PRP. The optimal eoncenlration uf PRP for promotion of the proliferation of BMSCs was 30%. There was no significant difference belween the aulologons amt allogeneic PRP (30%) regardiug their effects on proliferation and differentiation of BMSCs on day 4, 8, 12, 16 or 20 ( P 〉 0. 05) . On day 20, the cell apoptosis rates in blank group, conventional group, autologous group (30%) and allogeneic group (30%) were respectively 38.7% + 2.0%, 24. 1% -+ 1.7%, 9.7% + 0.8% and 10. 3 + 1.2%, with no significant differenc
