摘 要:利用抑制性消减杂交技术(SSH)分别对中国明对虾在卵黄发生的两个关键时期(卵黄发生前期和消退期)构建卵巢和肝胰腺的正反向消减文库,测序结果与GenBank进行BLASTx同源比较并利用GO分析进行功能注释。结果显示,各文库中的差异表达基因与卵巢发育时期相吻合,卵黄发生前期的卵巢和肝胰腺中分别检测到与卵黄蛋白原相关基因的出现,说明卵黄发生前期与卵黄发生有重要联系。在消退期中检测到与皮质棒形成相关的基因,说明消退期仍有皮质棒的形成,同时也检测到一些与免疫相关的基因,在机体中差异表达时期的功能有待进一步探讨。In the present study, the forward and reverse subtractive libraries of ovary and hepatopancreas were constructed in order to study the differentially expressed genes during two critical periods :previtellogenesis stage and declining stage of the shrimp Fenneropenaeus chinensis "s vitellogenesis. The sequencing results were compared with BLASTx homology on GenBank and conducted functional annotation via the Gene Ontology. The results showed that differentially expressed genes of subtractive library were in accordance with the ovarian development. The genes in relation to vitellogenesis detected in the hepatopancreas and ovarian during the previtellogenesis stage showed that previtellogenesis stage was the most important stage in the vitellogenesis; the genes in relation to the cortical stick formation were detected during the declining stage, which implies that the cortical stick was still forming during the declining stage. The other detected genes in relation to immune function would be further studied.
