摘  要:FJ2菌株为从土壤中筛选出的对马齿苋具良好除草活性的放线菌,对FJ2菌株发酵产物理化性质及除草活性物质的提取进行了研究。发酵产物理化性质研究结果表明:该活性物质对日光灯和自然光照射稳定,对70℃以下的高温相对稳定,对pH值在7~8范围内比较稳定,在4℃保存15d后,与对照差异不显著,且均无结块、分层现象。通过对FJ2菌株发酵液的预处理、旋转蒸发仪浓缩、溶媒萃取法、硅胶柱层析、TLC检测和大孔吸附树脂法等试验研究,明确了FJ2菌株除草活性物质初步分离纯化的技术流程。选择大孔树脂HP20进行吸附,以75%乙醇为洗脱剂动态解析初步提取FJ2菌株的除草活性物质获得粗品。经过硅胶柱用氯仿、乙酸乙酯和甲醇梯度洗脱,从粗品中分离得到了19个组分,其中活性最高的为第9组分,校正抑制率达86.93%。An Actinomycete strain FJ2 was isolated from soil which showed stronger herbicidal activity to Portulaca oleracea L. The fermentation physicochemical property and extraction herbicidal activity substances of strain FJ2 was studied. The results showed that the herbicidal substance was stable to natural light and fluorescent lamp. It was also relatively stable in The hyperthermia at about 70℃ and pH 7-8. It could be stored for 15d under 4℃, no significant difference with control, and there were no agglomerate, layer phenomenon. The technical process of herbicidal activity material separation of strain FJ2 was cleared by pretreatment, solvent extraction, silicagel column, TLC and macroporous adsorption resin methods. Macroporous resin HP20 was adopted to adsorb the herbicidal activity of strain FJ2, and 75% ethanol was selected for desorption the herbicidal activity with dynamic experiment. The crude antibiotic was well purified and 19 components were gained by this purification procedure. The ninth component was the best active component and its corrected inhibition rate reached 86.93%.
