摘  要:目的探讨人参皂苷Rg2后处理对心肌缺血再灌注损伤的保护作用及其机制。方法选择Wistar大鼠24只,分为空白处理组(A组)、模型组(B组),人参皂苷Rg2 50 mg/kg组(C组)及100 mg/kg组(D组),每组各6只。A组不予任何处理,B^D组均于结扎大鼠前降支动脉30 min后再灌注24 h,制备心肌缺血再灌注损伤大鼠模型,然后予药物灌胃,其中B组予生理盐水,C、D组分别予人参皂苷Rg2 50、100 mg/kg,连续7 d。计算心肌梗死面积,测定心肌组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量,逆转录PCR检测心肌内皮型-氧化氮合酶NOS(eNOS)水平。结果 B^D组心肌梗死面积、SOD活性、MDA、eNOS水平与A组比较差异均有统计学意义(p<0.05),而C、D组的心肌梗死面积小于B组,SOD活性增高,MDA、eNOS水平升高(p<0.05或p<0.01)。结论人参皂苷Rg2对实验性心肌缺血再灌注损伤有明显的保护作用,这可能与其抗氧化活性升高,自由基氧化损伤减少,eNOS增加有关。Objective Discuss the function and mechanism of Rg2 on myocardial ischemia -reperfusion in-jury in rats. Methods Choose 24 Wistar rats, divided it into normal control group ( Group A), model group ( Group B), Rg2 50 mg/kg group(Group C) and 100 mg/kg group( Group D), each group have six rats. Group A without any treatment, For Group B - D, after ligation descending branch of coronary artery 30 minutes, do reperfusion treat-ment on rats for 24 h so that preparing myocardial ischemia - reperfusion injury model. In next 7 days, administered some physiological saline to the rats of Group B and Rg2 50,100 mg/kg to the rats of Group C, D respectively. Then Calculate myocardial infarct size, determine superoxide dismutase (SOD) activity and malondialdehyde (MDA) in my-ocardial tissue, and determine myocardial endothelial - nitric oxide synthase NOS(eNOS) level by RT-PCR meth-od. Results Compared with Group A, the infarct size, SOD activity, MDA and eNOS level of Group B ~ D have sta- tistically significant differences(p 〈0.05). In group C and D, The myocardial infarct size is smaller, SOD activity and MDA,eNOS level is higher than group B(p 〈 0. 05 or p 〈 0. 01 ). Conclusion Ginsenoside Rg2 has significant protective effect on experimental myocardial ischemia - reperfusion injury, maybe because antioxidant activity in-crease, free radical oxidative damage reduced or eNOS increase in the organism.
