摘 要:目的:观察血必净注射液对系统性红斑狼疮(SLE)肾脏损害的可能干预效应。方法:采用30只清洁级EB病毒膜抗原BLLF1基因转基因小鼠为SLE动物模型,均为雌性,体重30~40 g。分为模型组(随机选择模型鼠)、8周龄给药组、10周龄给药组,每组10只。给药组分别给予6.4 ml/kg血必净注射液腹腔内注射,每日1次,连续14 d。然后无菌取血及肾脏,进行抗dsDNA抗体滴度及血清肾功能指标的检测,肾组织采用PASM染色进行组织病理学观察。远交系昆明种雌性小鼠10只作为正常对照组,于相应时间点取材观察上述指标的变化。结果:SLE小鼠抗dsDNA抗体滴度、血肌酐(Cr)和尿素氮(BUN)含量均明显高于正常对照组(P<0.05)。血必净注射液治疗能显著降低8周龄和10周龄SLE小鼠抗dsDNA抗体的滴度,显著降低其血Cr和BUN含量(P<0.05或P<0.01),并明显减轻狼疮鼠肾脏组织系膜基质的增生,但两不同周龄给药组间差异无显著性(P>0.05)。结论:血必净注射液治疗对SLE小鼠的肾脏损害具有防护作用。Abstract Objective:To evaluate the effect of Xuebijing injection ( 血必净注射液) on kidney injury in systemic lupus erythematosus (SLE)-prone BLLF1 mice. Methods: Ten normal female Kunming mice aged from 8 to 10 weeks old constituted the normal group (n= 10). A widely used mouse model, SLE-prone BLLF1 mice aged 8-10 weeks, were used as the experimental group. Thirty BLLF1 females were divided into 3 groups: SLE model group (5 8-week-old and 5 10-week-old BLLF1 mice chosen randomly, n= 10), 8-week-old SLE treatment group (n= 10), and 10-week-old SLE treatment group (n= 10). The two treatment groups received Xuebijing injection 6.4 ml/kg intraperito- neally once a day for 2 weeks. Kidney and serum samples were obtained from all animals. Levels of anti-dsDNA antibodies were observed, and serum creatinine (Cr) and blood urea nitrogen (BUN) levels were determined by biochemical method. Renal tissue sections were stained with Masson' s trichrome and periodic acid-silver methenamine. Histopatholog- ical changes in the kidney were evaluated by light microscopy. Results:The levels of anti-dsDNA, Cr and BUN were sig- nificantly higher in SLE model mice compared to the normal control group (P^0. 01). Treatment with Xuebijing injection could significantly decrease levels of anti-dsDNA antibodies, Cr and BUN (P%0. 05 or P〈0. 01). Pathological observation showed that the hyperplasia of renal mesangial matrix was alleviated in untreated BLLF1 mice. In contrast, minimal histopathological abnormalities were observed in the Xuebijing injection treatment group compared to the model group, but no difference was found between 8- and 10-week old treatment group(P〉0. 05). Conclusion:Treatment with Xuebijing injection can markedly attenuate renal injury in SLE-prone BLLF-1 mice.
