摘 要:目的探讨富含亮氨酸重复序列的G-蛋白偶联受体6(Lgr6)基因与人脐带间充质干细胞(hUC-MSCs)构建的组织工程细胞是否更快更好地修复小鼠皮肤创面。方法体外培养hUC-MSCs,构建pIRES2-EGFP-Lgr6表达载体,将表达Lgr6的质粒转染到hUC-MSCs中,然后种植于重症联合免疫缺陷小鼠背部全层皮肤烧伤创面(实验组),同时设种植转染空载质粒的hUC-MSCs作对照组。于术后7、14、21 d观察创面愈合率,并做HE染色和免疫组织化学。结果 hUC-MSCs能正确表达Lgr6靶蛋白,术后7、14、21 d实验组创面愈合率均明显高于对照组。HE染色显示,实验组新生表皮较厚,细胞层数较多且分层较明显,皮肤附属器增多,总体创面愈合情况均较对照组好;在再生表皮和真皮层中均发现有绿色荧光蛋白标记的细胞,且部分细胞表达人源性角蛋白14。结论 Lgr6基因转导入hUC-MSCs可应用于修复小鼠皮肤创面,修复效果较好。Objective To investigate the efficacy of tissue engineering leucine-rich repeat-containing G protein-coupled receptor 6(Lgr6) gene and human umbilical cord mesenchymal stem cells(hUC-MSCs) on skin wounds repairmen in mice.Methods Constructed pIRES2-EGFP-Lgr6 expressing vector was transfected into hUC-MSCs in vitro.The transfected hUC-MSCs were implanted on the back of full-thickness skin burn mice with severe combined immunodeficiency(SCID),and blank hUC-MSCs were applied in control group.On the 7th,14th and 21st day after implantation,the area percentage of wound healing was calculated,while HE staining and immunohistochemistry were performed for pathological observation.Results Lgr6 was successfully expressed in transfected hUC-MSCs.The area percentage of wound healing was significantly higher in experimental group than that in control group.Thicker skin tissues,more cell layers and stratification,increased skin appendages,and better overall wound healing was observed in experimental group than control group.Green fluorescent protein(GFP) cells were observed in the regenerating epidermis and dermis,some of which human-derived cytokeratin14(CK14) was expressed.Conclusion The hUC-MSCs expressing Lgr6 can be applied for skin wounds repairmen in mice,with better efficacy.
