摘 要:目的探讨原花青素(PA)对牙源性明胶酶/胶原酶活性的影响。方法选取新鲜离体牙本质,液氮冷冻下磨粉。将富含PA的葡萄籽提取物粉末加入乙醇溶剂中,制备不同浓度的PA预处理剂,用其预处理脱矿牙粉60、120s,蒸馏水反复离心冲洗,干燥。采用EnzChek试剂盒分析不同处理组牙粉内明胶酶/胶原酶活性。结果随着与试剂盒中底物反应时间的延长,各组牙粉明胶酶/胶原酶活性均逐渐升高。而无论预处理脱矿牙本质60s或120s,PA预处理组的牙源性明胶酶/胶原酶活性均低于未处理组,差异有统计学意义(P<0.05)。结论 PA能抑制牙源性明胶酶/胶原酶活性。Objective To evaluate the potential inhibitory effect of proanthocyanidins (PA) from grape seeds extract on the activities of endogenous gelatinase and collagenase in demineralized dentin matrix. Methods PA-based preconditioners were prepared by adding powdered PA-rich grape seeds extract to aqueous ethanol at various concentration. Extracted intact human third molars were collected, powdered in liquid nitrogen and pretreated by PA preconditioners at various concentration (15% PA or 10% PA) and for different pretreatment durations (60 s or 120 s). All the specimens were thoroughly rinsed by repeated eentrifugation, kept drying and lyophilized. EnzChek gelatinase/eollagenase assay was carried out to investigate the activities of gelatinase and collagenase in demineralized dentin matrix. Results Fluorescence attributed to the activities of gelatinase and collagenase gradually increased along the reaction. When dentin powder was pretreated by 10% PA, 15% PA (for either pretreatment duration), both the activities of gelatinase and collagenase were greatly inhibited in comparison to the non-treated control (P 〈 0.05). Conclusion PA pretreatment was remarkably effective in silencing the activities of endogenous gelatinase/collagenase in demineralized dentin.
