摘 要:目的观察耳聋左慈丸及其药对拆方对庆大霉素(GM)耳毒性模型的作用。方法出生2~3天小鼠的耳蜗螺旋器培养24h后,加入GM或同时加入GM和中药及其拆方的水提醇沉液处理。毛细胞经TRITC-phalloidin染色后在荧光显微镜下观察并进行全耳蜗毛细胞计数,计数结果输入全耳蜗毛细胞定量分析软件,获得全耳蜗毛细胞损失率。结果GM可造成耳蜗内、外毛细胞的缺失,且呈剂量依赖和时间依赖(P〈0.01);耳聋左慈丸全方可降低0.5mMGM造成的耳蜗内毛细胞(IHC)和外毛细胞(OHC)的损失率;耳聋左慈丸拆方补肾药对、养肝药对、利水药对和健脾药对均可降低0.5mM GM造成的耳蜗OHC的损失率(P〈0.05,P〈0.01),养肝药对和利水药对可降低IHC的损失率(P〈0.05)。结论耳聋左慈丸方中按照中医理论拆方配伍的药对(补肾药对、养肝药对、利水药对和健脾药对)对GM所致的OHC或IHC损伤有拮抗作用。Objective To observe the antagonistic action of "Erlong Zuoci Pill" and paired-medicine of it in rat models. Methods The organ of Corti explants from postnatal 2 - 3 day mouse ' s cochlea were Cultured cochlear spiral in postnatal 2 - 3 day mice for 24h, added the GM or GM and water extraction of deposed prescription. The hair cells were stained by TRITC-Phalloidin and counted by fluorescence microscope. Input the counts of cochlear hair cells into the full quantitative analysis software to get the full cochlear hair cell loss rate. Results GM can cause loss of outer hair cells (OHCs) and inner hair cells (IHCs) in a dose and time dependent manner ( P 〈 0.01 ) ; "Erlong Zuoci Pill" can reduce the loss of OHCs and IHCs caused by 0.5 mM GM ; paired medicine with nourish kidney, nourish liver, disinhibit water or strengthen spleen function can reduce the loss of OHCs caused by 0.5 mM GM ( P 〈 0.05, P 〈 0.01 ) , and paired medicine with nourish liver or disinhibit water function can reduce the loss of IHC ( P 〈 0.05 ). Conclusion The paired-medicine in "Erlong Zuoci Pill" has antagonistic action on 0HC or IHC caused by GM.
