摘 要:目的观察降浊化瘀合剂对2型糖尿病合并非酒精性脂肪肝大鼠的肝脏及大血管NF-κB通路的影响。方法采用改进高脂饲料喂养结合小剂量链脲佐菌素注射,建立大鼠模型,按体质量随机分为中药组、西药组和模型组,分别予降浊化瘀合剂、马来酸罗格列酮片和蒸馏水灌胃,并设正常组对照。给药8周后,测血清肿瘤坏死因子(TNF-α),Western-blot法检测肝脏和腹主动脉NF-κB(P65)及IκB蛋白表达。结果给药8周后,中药组较模型组大鼠血清TNF-α下降(P<0.05),并低于西药组(P<0.01)。与模型组比较,中药组大鼠肝及腹主动脉NF-κB(P65)蛋白表达下降(P<0.01),西药组大鼠肝脏NF-κB(P65)蛋白表达差异无统计学意义(P>0.05),西药组大鼠腹主动脉NF-κB(P65)蛋白表达降低(P<0.01);中药组均低于西药组(P<0.01)。与模型组比较,中药组肝脏IκB蛋白表达增强(P<0.05),而西药组差异无统计学意义(P>0.05),各组腹主动脉IκB蛋白表达差异均无统计学意义(P>0.05)。结论降浊化瘀合剂可明显抑制2型糖尿病合并非酒精性脂肪肝大鼠的肝脏和腹主动脉NF-κB通路。Objective To observe the effect of "Jiangzhuo Huayu Mixture" on NF-κB pathway of rats with type 2 diabetes and nonalcoholic fatty liver disease. Methods The rats models were established by fatty diet and small dose Streptozotocin Injection and randomly divided into three groups: Chinese medicine group was treated by "Jiangzhuo Huayu Mixture", western medicine group was treated by Roslglitazone and model group was treated by distilled water, and normal control group was established as well. After treated for 8 weeks, the TNF-α was observed and expression of NF-κB and IκB of liver and aorta abdominalis were detected by Western-blotting. Results After 8 weeks administration, the TNF-α in blood serum in Chinese medicine group was decreased ( P 〈 0.05 ) and lower than that of the western medicine group (P 〈 0.01 ). Compared with the model group, the expression of NF-κB (P65) of liver and aorta abdominalis in Chinese medicine group was decreased (P 〈 0.01 ), and no significant difference was found in NF-κB (P65) of liver between model group and western medicine group ( P 〉 0. 05 ) , expression of NF-κB( P65 ) of aorta in western medicine was decreased ( P 〈0. 01 ) ; the indexes in Chinese medicine group were lower than that of the western medicine group ( P 〈 0. 01 ). Compared with model group, the expression of IκB protein in liver of the Chinese medicine group was increased (P 〈 0.05 ), and no significant difference was found between western medicine group and Chinese medicine group ( P 〉 0.05 ). No significant difference was found in expression of IκB protein of aorta abdominalis among all the groups ( P 〉 0.05 ). Conclusion "Jiangzhuo Huayu Mixture" can obviously suppress the liver and abdominal aortic NF-κB pathway activation in rats with type 2 diabetes and nonalcoholic fatty liver disease.
