维A酸对肺气肿大鼠氧化/抗氧化因子表达调控作用的研究

摘  要:目的 探讨全反式维A酸(ATRA)对猪胰蛋白酶(PPE)诱导肺气肿大鼠模型氧化/抗氧化因子表达的调控作用.方法 选择60只SPF级健康Wistar大鼠,按数字表法随机分为对照组(N组)、模型组(E组)、棉籽油组(C组)、小剂量ATRA治疗组(L组)、中剂量ATRA治疗组(M组)和大剂量ATRA治疗组(H组)各10只作为研究对象,一次性气管内注入PPE制作大鼠肺气肿模型,然后选用小、中、大剂量ATRA及棉籽油注入大鼠腹腔内进行干预治疗.30 d后处死、解剖大鼠,测量大鼠肺体积、肺组织超氧化物歧化酶(SOD)活力和脂质过氧化物(MDA)含量.结果 与N组比较,E组、C组、L组、M组、H组的肺体积明显增大(t =9.351、7.805、2.155、2.273、5.507,均P<0.05),各组MLI、MAA的肺泡形态学比较显示:E组、C组、L组、M组、H组与N组,差异均有统计学意义(t=4.048、5.650、3.479、10.487、12.795,均P<0.05),L组比E组明显缩小(t =5.856,P<0.05);E组、C组、L组、M组、H组SOD活力与N组差异均有统计学意义(t=2.711、4.398、15.767、5.672、5.415,均P<0.05),L组SCD活力明显高于其他各组(t=15.767、13.502、12.139、7.866、7.221,均P<0.05).E组、C组、L组、M组、H组MDA含量均明显高于N组(=7.702、8.507、3.186、7.950,均P<0.05),L组、M组明显低于E组(t=6.397、4.044,均P<0.05).结论 采用低剂量ATRA治疗对肺泡有较好修复作用,可提高肺组织SOD活力,能够作为抗氧化作用保护剂,达到保护肺组织作用.Objective To observe the effect of all trans retinoic acid (ATRA)on the oxidative stress influ- ence in emphysema model rats induced by porcine pancreatic elastase (PPE). Methods 60 rats were randomly divid- ed into control(N) ,model group(E) ,cottonseed oil group(C) ,low dose of ATRA treatment group(L) ,medium dose of ATRA treatment group (M) and high dose of ATRA. treatment group (H). The rat models of emphysema were con- structed by a single intratracheal instillation of PPE and treated with low, medium and large dose of ATRA and cotton- seed oil by intra-abdomirtal injeetior, daily. 30 days later, rats lung volume, the alveolar morphology, lung tissue SOD and MDA content were measured. Results The lung vllume of E, C, L, M, H group were significantly greater than that of N group, The alveolar morphological and lung tissue oxidative stress of E, C, L, M, H group were also significant differences compared with N group ( P 〈 0.05 ), and those of L group were significantly decreased compared with E group. Lung tissue SOD achvity and MDA content of E, C, L, M, H group were significantly higher than N group (P 〈 0.05). SOD activity of L group was higher than that of others groups ( P 〈 0.05 ). MDA content of L, M group were lower than E group ( P 〈 0.05 ). Conclusion Emphysema rat models with low dose of ATRA therapy on alveolar have better repair function,which can protect lung tissue by improving lung tissue SOD and reducing lung tissue MDA con- tent.
