不同保存期库血辐照前后的淋巴细胞代谢活性研究

摘　　要：目的:了解不同保存期内库血辐照前后的淋巴细胞活性变化。方法:采用经典的MTT实验法,检测4℃保存1、5、10、15d内的库血在辐照前后淋巴细胞的增殖代谢活性,以未辐照血为对照组,根据公式计算辐照的抑制率并绘制曲线。结果:辐照对在4℃保存5d以内的库血表现出增殖抑制效应,但淋巴细胞在辐照后仍保持一定的活性,代谢抑制率为45.26%;保存10d以上的库血,辐照的抑制效应不明显,仅为10%左右。结论:随着库血在4℃保存时间的延长,辐照对淋巴细胞增殖的抑制效应递减。Objective：To evaluate the metabolism activity of lymphocytes in different storage period before and after irradiation.Method：The metabolic activity of lymphocytes separated from blood which preserved in 1,5,10 and 15 days in 4℃ was detected by using the cell viability MTT assay.Non irradiated blood was used as control group.A curve was draw by calculating the radiation inhibition rate according to the formula.Result：Proliferation inhibiting effect was showed on lymphocytes preserved within 5 days which could maintain certain activity and the metabolic inhibition rate was 45.26%.Inhibition effect of irradiation was not obvious on lymphocytes preserved more than 10 days.Conclusion：Inhibition effect of irradiation on the lymphocyte proliferation would diminishes as the time of blood storage.
