摘 要:目的:了解不同保存期内库血辐照前后的淋巴细胞活性变化。方法:采用经典的MTT实验法,检测4℃保存1、5、10、15d内的库血在辐照前后淋巴细胞的增殖代谢活性,以未辐照血为对照组,根据公式计算辐照的抑制率并绘制曲线。结果:辐照对在4℃保存5d以内的库血表现出增殖抑制效应,但淋巴细胞在辐照后仍保持一定的活性,代谢抑制率为45.26%;保存10d以上的库血,辐照的抑制效应不明显,仅为10%左右。结论:随着库血在4℃保存时间的延长,辐照对淋巴细胞增殖的抑制效应递减。Objective:To evaluate the metabolism activity of lymphocytes in different storage period before and after irradiation.Method:The metabolic activity of lymphocytes separated from blood which preserved in 1,5,10 and 15 days in 4℃ was detected by using the cell viability MTT assay.Non irradiated blood was used as control group.A curve was draw by calculating the radiation inhibition rate according to the formula.Result:Proliferation inhibiting effect was showed on lymphocytes preserved within 5 days which could maintain certain activity and the metabolic inhibition rate was 45.26%.Inhibition effect of irradiation was not obvious on lymphocytes preserved more than 10 days.Conclusion:Inhibition effect of irradiation on the lymphocyte proliferation would diminishes as the time of blood storage.
