摘 要:目的开发新型细胞保存固定液并探讨其对子宫颈炎样本的液基细胞学镜检效果的影响。方法新型细胞保存固定液由溶血剂、固定剂以不同体积比(1 ∶10~1 ∶100)配制而成,其中溶血剂为含0.8%SDS、1.7%NH4Cl、0.9%NaCl水溶液,固定剂则为甘油 ∶乙醇=1 ∶2组成的混合液。用25%苯酚胶浆诱导大鼠子宫颈炎模型,造模9天后,用棉签刮取子宫颈口细胞,将其分别浸入传统细胞保存固定液(1.2%草酸铵溶于95%乙醇)及新型保存固定液中,离心、制片、巴氏染色后进行镜检。结果传统保存固定液组的涂片含有较多的红细胞及黏液,被检细胞成团,形态欠清晰;新型保存固定液组的涂片细胞分布均匀,被检细胞形态清晰可见,无红细胞及黏液残存,其整体镜检效果具有明显优势,且新型细胞保存固定液中溶血剂和固定剂的配比为1 ∶40时,所制涂片的镜检效果最佳。结论新型细胞保存固定液保存固定子宫颈细胞的效果明显优于传统细胞保存固定液,溶血剂和固定剂的适宜配比为1 ∶40。Purpose To establish a new cell-preserving liquid and to investigate its application for the elimination of red blood cells in the liquid-based specimens of eervicitis. Methods The new cell-preserving liquid was prepared according to different molar ratio of hemolytic agent and fixative. The hemolytic agent contained 0.8% SDS and 1.7% ammonium chloride dissolved in normal saline, the fixative was uniformly mixed with glycerol and anhydrous ethanol in the molar ratio of 1 : 2. Nine days after vaginal administration of phenol gelatinoids in rats, the cervical cells from rats were scraped by cotton swab, and were immersed in one group of traditional cell preservation liquid ( 1.2% ammonium oxalate dissolved in 95% ethanol) and other six groups of novel cell preservation liquid, respec- tively. After the centrifugation, the cells in each container were resuspended in 20 p,1 cell preservation liquid, followed by placing onto a polylysined slide. For morphological observations the cells were stained using the papanicolaou technique. Results Morphological observations indicated that the cellular distribution of smears using novel cell preservation liquid was uniform well with disappearance of red blood cells, and the cellular morphology exhibited clearly, in comparison to traditional preservation liquid group. Moreover, the smears exhibited the best effect under the combination of the hemolytic agent and the fixative in a molar ratio in the range 1 : 40. Conclusion These findings demonstrate that the novel cell preservation liquid displays a better effect on the preservation of cervical cells with the combination of hemolytic agent and fixative in a molar ratio in the range 1 : 40.
