生脉散对db/db小鼠心肌组织病理变化的影响

摘　　要：目的:探讨生脉散对糖尿病心肌病小鼠心肌组织病理变化的影响。方法:将8周龄db/db小鼠随机分为模型组和生脉组,另设背景系小鼠C57BLKS/J为正常组。生脉组给予4.5g·kg-1·d-1生脉散灌胃,干预24周后检测小鼠空腹血糖、体质量、心脏质量、血流动力学指标,并以HE染色及透射电镜观察心肌组织形态变化、Masson染色观察心肌组织胶原纤维。结果:与正常组比较,模型组db/db小鼠空腹血糖、体质量、心脏质量及心肌胶原容积分数(CVF)显著升高(P<0.05),左室内压最大下降速率(-dp/dt)明显下降(P<0.05);生脉组db/db小鼠心脏质量与CVF较模型组明显减轻(P<0.05),而-dp/dt较模型组明显增加(P<0.05)。组织学检查显示,模型组db/db小鼠存在心肌细胞肥大、室壁增厚、间质纤维化、微血管病变及线粒体损伤,生脉组上述变化有一定程度的改善。结论:生脉散对db/db小鼠心肌损伤具有保护作用。Objective ; To study the effects of “ Shengmai Powder” on myocardium in db/db Mice. Methods ： Thirty db/db mice aged 8 weeks were randomly divided into model group （n = 15） and “ Shengmai Powder” group （n = 15 ）. While 15 C57BLKS/J inbred mice at the same age were divided into control group. 4. 5 g ？ kg^-1 ？ d^-1 “ Shengmai Powder” was intragastrically administrated to those mice in “ Shengmai Powder” group. After 24 weeks, the blood glucose, body weight and heart weight, haemodynamics were determined. The myocardial structure and sub-cellular structures changes were observed by HE staining and electron microscopy. Results： Compared with the control group, the blood glucose, body weight, heart weight and the CVF in model group were significantly increased （P 〈0. 05 ）,and the - dp/dt was significantly decreased （ P 〈 0. 05） ; the heart weight and the CVF in “ Shengmai Powder” group were decreased and the - dp/dt was increased significantly （P 〈 0. 05）. The cardiomyocyte hypertrophy, interstitial fibrosis, microvessel basement membrane thickening and mitochondrial swelling were improved in model “ Shengmai Powder” group. Conclusion： “ Shengmai Powder” has positive effect on cardiac hypertrophy and myocardial fibrosis in db/db mice.
