摘 要:目的 从草乌中分离草乌多糖 ,并研究其组成性质。方法 采用DEAE纤维素 (DEAE 5 2 )柱层析和凝胶柱层析分离纯化草乌多糖 ,气相色谱法和薄层层析法测定其多糖组成 ,凝胶过滤法测定其相对分子质量 ,红外光谱测定其苷键类型 ,比色法测定其糖含量。结果 分离得到的草乌多糖 (AconitumkusnezoffiiReichb .,AK)含有鼠李糖、木糖、甘露糖、葡萄糖、半乳糖、阿拉伯糖 ,它们的摩尔比为 1∶1.7∶2 0 3∶1.7∶5 9.7∶3 .5。其糖含量 88%。AK的平均相对分子质量为 2 .8× 10 4。它的红外光谱中有典型的多糖吸收峰 ,并具有α 型糖苷键 ,紫外扫描无核酸和蛋白特征吸收峰。结论 AK首次从草乌块根中提取获得 。OBJECTIVE To study the isolation and composition of Aconitum kusnezoffii Reichb.polysaccharkide(AK).METHODS AK was purified by DEAE ion exchange cellulose(DEAE 52) and sepharose Cl 4B.The component of the saccharide was determined by GC and TLC.Its MW was measured by gel filtration.The form of glucosidic bond was detrermined by IR.The content of the saccharide was measured by colorimetry.RESULTS Rhamnose,xylose,mannose,glucose,galactose and arabinose were included in AK with the molar ratio of 1∶1.7∶203∶1.7∶59.7∶3.5.Its average MW was estimated t be 2.8×10 4.The content of saccharide was 88%.Typical absorption of polysaccharide was shown in its IR spectrum.It contained α glucosidic bonds by IR analysis.It had no typical absorption of nucleic acid or protein by UV scanning.CONCLUSION AK is first isolated from the root of Aconitum kusnezoffii Reichb..
