摘  要:目的 研究环境雌激素己烯雌酚(DES)作用于孕鼠,对子鼠生长发育过程中睾丸组织黄体生成素(LH)及其受体(LHR)表达的影响。方法怀孕雌性昆明小鼠200只,随机分成8组,实验组1.6分别给予DES0.1、0.5、10.0、25.0、50.0及100.0μg/kg·d-1。DES以DMS0和生理盐水为溶剂。对照组仅给予DMSO和生理盐水;正常组不给任何药物。新生小鼠出生后,以小鼠出生第1天记为0,分别于0,5,15,25,35,45,60d处死6只雄性小鼠并取其睾丸,常规固定、制片,应用免疫组织化学方法检测睾丸组织中LH和LHR的表达。结果中、高剂量(DES10.0、25.0、50.0及100.0ug/kg·d。组)的实验组中LH和LHR表达明显降低(P〈0.05),均有显著的剂量效应关系。而低剂量实验组(DES0.1和0.5Pg/kg·d-1组)则变化不明显(JD〉0.05)。结论中高剂量DES可抑制睾丸LH及其受体的表达,有剂量效应关系,从而影响生殖功能。低剂量DES则无反应。Objective To study the effects of prenatal exposure to diethylstilbestrol (DES) on the expressions of LH and LHR in testica tissue of offspring male mice. Methods Pregnant mice were randomly divided into 8 groups and injected with DES subcutaneously from the 9th gestational day to the 17th day at dosages of 0.1, 0.5, 10.0, 25.0, 50.0 and 100.0μg/kg·d-1 dissolved in dimethyl sulfoxide (DMSO), and mice in the normal group were not given any drug, mice in the control group were given DMSO and 9%NS, The day of birth was designated as day 0. Male offspring mice were sacrificed on Days 0, 5, 15, 25, 35, 45and 60, and their testical tissues were collected for histological analysis. The expressions of LH and LHR in testis were detected by immunohistochemistry. Results Expressions of LH and LHR in testis were lower in DES-treated groups (DES10.0, 25.0, 50.0 and 100.0μg/kg.d1 group) than those in the normal and control groups (P〈0.05). But there were no significant differences between low-dose treated groups (DES0.5 and DES0.1μg/kg.d-1 group) and the normal or control group. Conclusion The expressions of LH and LHR in the offspring male mice' s testis had no significant differences when the pregnant mice exposured to Low-dose DES. High-dose DES can inhibit the expressions of gonadotrophins and their receptors in offspring male mice's testis.
