摘  要:为研究纳米银体外抗新城疫病毒(NDV)的作用,并初步探讨其作用机制,运用细胞培养技术和MTT方法,分析纳米银对NDV感染HeLa细胞的预防、治疗作用以及对NDV的直接灭活作用;通过血凝抑制试验和神经氨酸酶活性抑制试验,观察纳米银对血凝素—神经氨酸酶活性物质的影响;采用透射电镜负染技术观察纳米银对新城疫病毒形态结构的直接影响。结果显示:NDV可通过不同途径感染HeLa细胞,纳米银组的细胞存活率与NDV对照组比较有明显的统计学差异(P<0.01);在体外将NDV和纳米银作用不同时间后,NDV血凝效价明显降低;纳米银对NDV神经氨酸酶活性的抑制率均高于80%,而病毒对照组和溶剂组的抑制率均低于20%;透射电镜观察表明,纳米银对NDV形态结构的破坏作用随着时间的延长而增加。结果表明,纳米银在体外对NDV具有明显的抑制作用,其抑制机制可能与纳米银破坏病毒粒子结构及抑制神经氨酸酶活性有关。To explore the inhibiting effect and mechanism of silver nanoparticles against Newcastle disease virus (NDV), cell culture and MTT test assay were applied to evaluate anti-NDV activitiy of silver nanoparticles,hemagglutinin inhibition test and neuraminidase activity inhibition test were used to study the inhibitory effect of silver nanoparticles on hemagglutinin-neuraminidase activity and destructive effect of silver nanoparticles on NDV was observed by transmission electron microscope (TEM). The resuhs showed that under three different ways, the survival rates of cells in silver nanopartieles treatment group was statistically significant compared with NDV control group (P〈0.01). Silver nanoparticles were mixed with NDV in different time,the hemagglutination titer decreased obviously. The inhibition rate of neuraminidase activity of silver nanoparticles treatment group was higher than 80%,while NDV control group was lower than 20%;TEM showed that silver nanoparticles had a obvious destructive effect on NDV with a time-dependence trend. The results indicated that silver nanoparticles had remarkably inhibiting effects on NDV in vitro and the possible mechanisms could be associated with neuraminidase inhibition and virion damage by silver nanoparticles.
