摘  要:目的研究microRNA-328(miR-328)和microRNA-155(miR-155)在急性冠脉综合征(ACS)患者血浆中的水平及其在ACS诊断和鉴别诊断中的价值。方法收集56例ACS患者[其中24例为不稳定心绞痛(UA),32例为急性心肌梗死(AMI)]和20例体检健康者,分析两组一般临床资料及相关生化指标;用实时荧光定量聚合酶链式反应(qRT-PCR)检测两组研究对象血浆miR-328及miR-155的含量,并分析其与心肌肌钙蛋白T(cTnT)的相关性。结果 ACS组与健康对照组血浆miR-328为(8.12±3.81 vs 1.00±0.00),差异有统计学意义(t=7.924,P<0.05);AMI亚组与UA亚组血浆miR-328含量为(10.69±2.17 vs 4.26±2.06),差异有统计学意义(t=7.227,P<0.05)。ACS组与健康对照组血浆miR-155含量为(1.00±0.26 vs1.00±0.00),AMI亚组与UA亚组血浆miR-155含量为(1.02±0.28 vs 0.96±0.22),差异均无统计学意义(P>0.05)。血浆miR-328水平与cTnT呈正相关(r=0.434,P<0.05);ACS患者血浆miR-155水平与cTnT无相关性(r=-0.233,P>0.05)。结论 miR-328在ACS患者血浆中水平升高,可能作为ACS诊断及鉴别诊断的潜在生物标志物。Objective:To investigate the levels of blood plasma microRNA-328 (miR-328) and microRNA-155 (miR-155) in acute coronary syndrome (ACS) patients, and their values in the diagnosis and differential diagnosis of ACS. Methods:Twenty healthy volunteers and 56 ACS patients, including 24 with unstable angina (UA) and 32 with acute myocardial infarction (AMI), were enrolled, and their general clinical data and correlated biochemical parameters were analyzed. The levels of blood plasma miR 328 and miR-155 in all subjects were detected by real-time quantitative polymerase chain reaction (qRT-PCR), and their correlation with cardiac troponin T (cTnT) was analyzed. Results:There was significant difference of blood plasma miR-328 level between the ACS group (8.12±3.81) and the control (1.00±0.00) (t=7.924, P〈0.05). Moreover, the level of plasma miR-328 in AMI subgroup (10.69±2.17) was significantly higher than that in UA (4.26±2.06) (t=7.227, P〈0.05). However, there was no significant difference of plasma miR-155 level between the ACS and the control (1.00±0.26 vs 1.00±0.00, P〉0.05) and between the AMI and the UA (1.02±0.28 vs 0.96±0.22, P〉0.05). There was positive correlation between plasma miR-328 level and cTnT (r=0.434, P〈0.05), but no correlation between plasma miR-155 level and cTnT (r=-0.233, P〉0.05). Conclusion:There was higher plasma miR-328 level in the patients with ACS, indicating that plasma miR-328 may be used as a novel biomarker for the diagnosis and differential diagnosis of ACS.
