荭草苷对心肌梗塞大鼠的治疗作用研究

摘　　要：目的探讨荭草苷对心肌梗塞大鼠的治疗作用。方法建立冠脉结扎致大鼠心肌梗塞模型,将大鼠随机分为假手术组、模型组、阳性药白藜芦醇组及低、中、高剂量荭草苷组(1.0,2.0,4.0 mg/kg);采用多道生理记录仪采集心电图、血流动力学指标,湿重法记录心脏重量以及左心室重量;HE染色观察心肌组织病理学改变。结果与模型组比较,荭草苷能明显改善心肌梗塞大鼠心电图、血流动力学指标、心室重构及心肌病理损伤程度,而且剂量在4.0 mg/kg以内,疗效呈剂量依赖性增强。结论荭草苷对大鼠心肌梗塞具有明显治疗作用。Objective To investigate the therapeutic effects of orientin on myocardial infarction （MI） rats. Methods The rat MI models were established by ligating of the left anterior descending coronary artery. Twenty - four hours later after modeling, the survival model rats were randomly divided into MI group （ Model group, n = 12 ）, resveratrol group（ 2.5 mg/kg, n = 7 ）, orientin low dosage group （ 1.0 mg/kg, n = 8 ）, orientin middle dosage group （ 2.0 mg/kg, n = 10 ）, and orientin high dosage group （ 4.0 rag/ kg, n = 7 ）, with another sham - operated group （ Sham group, n = 11 ） as control. After 3 weeks, electrocardiogram （ECG） and hemodynamic index were recorded by PowerLab, the left ventricular weight index（LVWI） and lung weight were calculated. The tissue sample was taken from the left ventricle wall and was stained with hematoxylin and eosin （HE） and examined with a light microscope. Results 0rientin can ameliorate ECG changes, hemodynamics indexes, ventricular remodeling, myocardial structures in MI rats in a dose - dependent manner. Conclusion Orientin has the obvious therapeutic effects on MI rats.
