摘 要:目的探讨左归降糖方对高脂饮食2型糖尿病转基因MKR小鼠脑组织NF-κB信号通路炎症因子的影响。方法30只MKR小鼠随机分为MKR组、MKR高脂组和左归降糖方组;高脂饲料饲养MKR鼠加重体内糖脂代谢紊乱,左归降糖方组干预治疗高脂饮食MKR鼠4周,测定小鼠空腹血糖、血清胰岛素和血脂浓度,RT-PCR检测小鼠脑组织IKB、TNF-α和MCP-1基因mRNA表达变化。结果与普通MKR小鼠比较,高脂饮食MKR小鼠空腹血糖、血清胰岛素、总胆固醇水平升高,甘油三酯水平下降,脑组织TNF-α和MCP-1基因mRNA表达水平均增加;左归降糖方降低高脂饮食MKR小鼠空腹血糖、血清胰岛素、总胆固醇以及甘油三酯水平,同时降低脑组织TNF-α、MCP-1基因mRNA表达水平。结论左归降糖方可通过抑制高脂饮食MKR小鼠脑组织NF-κB信号通路的激活来降低炎症损伤,对脑组织具有保护作用。Objective To observe the effects of Zuogui Jiangtang Recipe (ZJR) on inflammatory factors of NF - κB signaling pathway in the type 2 diabetes MKR mice raised by high fat feeds. Methods Thirty MKR mice are randomly divided into the control group (CG), the model group (MG) and the ZJQR group (ZG). Except for CG mice which are raised by standard animal feeds, others are both raised by high fat feeds to exacerbate disorders of glycometabolism and lipid metabolism. Then ZG mice have been administered by ZJQR for 4 weeks. Fasting blood -glucose (FBG), fasting serum insulin(FINS) , triglyceride (TG) and total cholesterol(TC) are examined, and RT- PCR is used to analyze the gene expressions of IKB, TNF-α and MCP- 1 in the brain of mice. Results The gene expressions of TNF - α and MCP - 1 on mRNA level in MG mice are elevated compared with CG, and so are FBG , FINS,TC ,while TG is decreased significantly ;ZJQR decreases the gene expressions of TNF-α and MCP- 1, and it decreases FBG , FINS,TG and TC, too. Conclusion ZJQR can reduce the infalmmation injury in the brain of MKR mice through controling the activation of NF - κB signaling pathway.
