摘  要:目的观察脑脉通对脑缺血再灌注大鼠移植BMSCs后神经营养因子表达变化的影响。方法体外培养和扩增BM-SCs;线栓法制备脑缺血模型;大鼠脑缺血再灌注后24 h颈内动脉移植BMSCs;免疫组织化学法测定BMSCs移植后7 d(早期)、14 d(中期)和28 d(后期)神经细胞特异性标志NSE和GFAP、神经营养因子NGF和GDNF表达变化。结果假手术组大鼠NSE和GFAP表达明显。脑缺血后7d、14d、18d组NSE表达减弱;7d组GFAP表达增强、14d和28d减弱;7d组NGF表达增强、14d和28d组递减;7d组GDNF表达增强、14d减弱、28d增强。脑脉通7d组大鼠的NSE表达较模型组增强;28d组GFAP表达减弱;14d和28d组NGF表达增强;7d组GDNF表达减弱,14d组增强。移植28d组GFAP,NGF和GDNF表达均较模型组增强。与移植组比较,联合7d组和28d组NSE表达增强;7d组GFAP表达减弱,14d增强;28d组NGF表达增强。结论脑缺血再灌注后神经元细胞反应、神经营养因子NGF和GDNF表达呈不同变化特点;BMSCs移植后早期作用并不显著,但后期增强神经营养因子表达及神经细胞反应的作用明显;脑脉通可使BMSCs移植保护神经细胞的作用提前并增强,其作用机制可能与早期上调NGF和GDNF表达有关。Objective To observe the changes of neurocytes and neurotrophic factors expression after being transplanted with bone mesenchymal stem cells (BMSCs)in rats with cerebral ischemia and the influence of Naomaitong (NMT) on the changes. Methods Rats whole bone marrows were cultivated and BMSCs were purified and increased in vitro. Middle cerebral artery occlusion (MCAO) model was duplicated with nylon thread. Rats of transplantation and combination groups were transplanted with BMSCs via carotid artery at 24 h after cerebral ischemia reperfusion. Expression of NSE and GFAP were determined by using immunohistochemical method at 7d( earlier period), 14d(metaphase) and 28d(auaphase) after BMSCs transplantation, as well as the expression of NGF and GDNF. Results Ratg expressions of NSE and GFAP were significant in sham operation group. NSE expression weakened after injury of cerebral ischemia. GFAP expression in 7d model group increased, while decreased in 14d and 28d model groups. Expressions of NGF and GDNF all enhanced in their 7d model groups, while NGF expression decreased progressively in 14d and 28d model groups. GDNF expression attenuated in 14d model group and increased in 28d group. In NMT groups, NSE expression increased and GFAP expression decreased in 7d groups, while GDNF expressions decreased in 28d group. GFAP, NGF and GI)NF expression all increased in 28d transplantation groups. In comparison with that of transplantation groups,NSE expression all increased in 7d and 28d combination groups, and GFAP expression decreased in 7d group and increased in 14d group. NGF expression increased in 28d group obviously. Conclusion It was showed that the expressions of neurotrophic factors performed different characteristics respectively after cerebral ischemia reperfusion. BMSCs transplantation showed no significant effects on the changes of neuron and neurotrophic factors in earlier period, while in metaphase and anaphase, the roles in increasing neurotrophic factors and neurocytes reaction
