摘  要:目的:观察虎杖水提物的抗炎作用,并从影响一氧化氮生成探讨其机制。方法:采用二甲苯致小鼠耳廓肿胀法和鸡蛋清致大鼠足跖肿胀模型观察虎杖水提物的抗炎作用;用脂多糖(LPS)刺激原代小鼠腹腔巨噬细胞,观察虎杖水提物对其iNOS mRNA的表达以及一氧化氮生成的影响。结果:虎杖水提物低剂量与高剂量均能明显减轻二甲苯所致的小鼠耳廓肿胀和鸡蛋清致大鼠足跖肿胀。虎杖水提物能明显降低LPS诱导的巨噬细胞iNOS mRNA的水平以及在质量浓度为10 mg.mL-1时显著减少其一氧化氮的生成。结论:虎杖水提物具有明显的抗炎作用,并抑制炎性因子一氧化氮的产生,可能是其减轻炎症反应机制之一。OBJECTIVE To observe the anti-inflammatory effect of the aqueous extracts from Rhizoma Polygoni Cuspidati (RPC) and its effect on production of nitric oxide (NO). METHODS The auricle edema model was induced by xylene in the mice and rat paw edema model was induced by injecting of egg white to investigate the anti-inflammatory effect of RPC aqueous extracts. Lipopolysaccharide (LPS) induced mouse peritoneal macrophages were used, then NO concentration in supernatant was measured by NO kit and the expression of iNOS mRNA in macrophages was detected by RT-PCR to study the effect of RPC on NO production and iNOS mRNA level in LPS-induced mouse macrophages. RESULTS Both low-dose (4 g,kg-1 ) and high-dose (8 g, kg-1) RPC aqueous extracts could reduced the ear edema induced by xylene in mice and paw ankle swelling in- duced by egg white in rats. The level of iNOS mRNA in LPS-induced mouse macrophages was remarkably decreased by RPC a- queous extracts at concentration of both 2 mg.mL -1 and 10 mg.mL -1 but NO production was inhibited only at 10 mg.mL-1. CONCLUSION The aqueous extracts of RPC have anti-inflammatory effect and the underlying mechanism may be related to its suppression of production of pro-inflammatory cytokine NO.
