层粘连蛋白和Ⅳ型胶原在小鼠肾泌尿小管发育中的表达

摘　　要：目的观察层粘连蛋白(LN)和Ⅳ型胶原(CoⅣ)在小鼠肾泌尿小管发育中的表达规律,探讨两者与肾泌尿小管发育的关系。方法选取胚龄(E)12、14、16、18d胎鼠和生后(N)3、7、14和40d仔鼠肾脏,应用免疫组织化学技术对小鼠肾泌尿小管LN和CoⅣ的表达进行定位观察;应用蛋白印迹技术对小鼠肾泌尿小管LN和CoⅣ的表达进行定量检测。结果 E12d,LN在输尿管芽上皮细胞基膜处开始微弱表达,而CoⅣ在输尿管芽无阳性表达;E14d,LN和CoⅣ在各期肾小管上皮细胞基膜处表达;E16d,LN和CoⅣ在集合管上皮细胞基膜处均有表达。随着肾脏的发育,LN和CoⅣ在肾泌尿小管的表达量逐渐增加。结论 LN和CoⅣ可能对小鼠肾泌尿小管发育和成熟起重要作用。OBJECTIVE To observe the expression rules and sites of LN and ColV in developing uriniferous tubules of mouse kidney and investigate the relationship between the expression and uriniferous tubules development. METHODS Immunohis- toehemistry and Western blot were used to observe and measure the expressions of LN and Co 1V in embryos of 12, 14, 16, 18 d and 3, 7, 14, 40 d postnatal mouse uriniferous tubules. RESULTS The expression of laminin was noticed in the ureteric bud in E12d, Coiv was not found in ureteric bud; in E14d and E16d, LN and CoIVall were found in all stages of renal tubules and collecting ducts, and increased gradually with ages; the expressions of LN and Co 1V were increased. CONCLUSION LN and CoiV might play an important role in the development and maturation of mouse uriniferous tubules.
