摘 要:采用子房注射法成功地将外源DNA导入五唇兰。结果表明:外源DNA在转化植株成熟果实中种子的GUS染色率达6.7%,无菌播种后经潮霉素筛选,获得18株幼苗,每株幼苗切取幼叶及幼根进行GUS检测,均为阳性;随机挑选7株幼苗进行PCR检测,均能扩增出GUS基因和潮霉素抗性基因条带,说明外源基因已经整合到植株基因组内。子房注射法在兰科植物转基因中的成功应用,可为兰科植物育种提供一种简单高效的途径。The method of ovary injection was first reported for Doritis pulcherrima Lindl. transformation in this study. Results showed that the GUS staining rate of seeds from the mature fruit by ovary-injection was 6.7%. Eighteen hygromycin-resistance plants were gained after screening. Seven plants selected randomly were detected by GUS assay and PCR analysis of GUS and Hyg, all of them were positive. The results showed the integration and expression of foreign gene in hygromycin-resistanee plants. The successful application of ovary injection for D. pulcherrima transformation provides a simple and efficient method for the Orchidaceae breeding.
