缺氧对胎盘分泌sFlt-1的影响

摘　　要：目的通过检测在不同氧浓度条件下培养的正常胎盘组织及妊娠期高血压疾病(hypertensive disorders of pregnancy,HDP)胎盘组织中可溶性血管内皮生长因子受体-1(soluble fms-like tyrosine kinase-1,sFlt-1)的分泌情况,从而了解缺氧因素对sFlt-1分泌的影响。方法将正常胎盘组织及HDP胎盘组织在正常氧浓度(20%氧浓度)及低氧浓度(1%氧浓度)条件下进行培养,通过检测人绒毛膜促性腺激素(human chorionic gonadotrophin,HCG)浓度了解胎盘组织分泌状态,从而筛选出培养生长良好的胎盘组织进行sFlt-1浓度检测,比较正常胎盘组织及HDP胎盘组织在正常氧浓度(20%氧浓度)及低氧浓度(1%氧浓度)条件的分泌浓度。结果对正常胎盘,正常氧浓度下第4天及第5天sFlt-1浓度分泌无差异,缺氧条件下sFlt-1分泌受抑制;对HDP胎盘,正常氧浓度下第5天较第4天sFlt-1分泌持续增加,缺氧条件下sFlt-1分泌受抑制。结论 HDP胎盘组织sFlt-1分泌的负反馈机制失灵;缺氧对正常或HDP胎盘组织的sFlt-1分泌起到抑制作用。Objective To study the influence of hypoxic condition on soluble fms - like tyrosine kinase - 1 （sFlt- 1 ） secretion by testing the sFlt - 1 secretion in normal placenta tissues and hypertensive disorders of pregnancy（HDP） placenta tissues under different levels of oxygen concentration. Methods Normal placenta tissues and HDP placenta tissues under normal （20%） and low （1%） oxygen concentration levels were cultured respectively. Chosen those placenta tissues in good condition by testing concentration of HCG, and their sFh - 1 concentration were measured. In different cases, the sFlt - 1 concentration of normal placenta tissues and HDP placenta tissues under normal （ 20% ） and low （ 1% ） oxygen concentration level were compared. Results For normal placenta tissues, there was no difference in sFh - 1 concentration on the 4th and 5th day under normal oxygen concentration. The sFh - 1 concentration of HDP placenta tissues kept rising on the 5th day comparing to the 4th day under normal oxygen concentration, but it kept failing on the 5th day comparing to the 4th day under low oxygenconcentration. Conclusion The negative feedback mechanism of sFlt - 1 secretion is not observed in HDP placenta tissues. The hypoxic condition suppresses the sFlt - 1 secretion in both normal and HDP placenta tissues.
