中药方剂对切割海马伞大鼠学习、记忆的保护作用

摘  要:目的 :研究以党参、黄芪、何首乌、山萸肉等益肾健脑、益气养血为主方的中医药对切割海马伞大鼠学习、记忆力的影响以及对隔区胆碱能神经元的保护作用。方法 :实验组和对照组动物切割海马伞前后分别用中药煎剂和自来水灌胃 ,术后第 3、4周行避暗回避试验和跳台试验 ,最后灌注固定动物 ,取隔区切片作 Ch AT免疫组化和 NOS组化染色 ,显微镜下记数切割海马伞侧内侧隔核和斜角带垂直支 Ch AT和 NOS阳性神经元数 ,并观察胞体和纤维变化。结果 :避暗回避试验的探索次数、滞留时间及跳台试验的主动和总回避反应阳性率实验组均好于对照组 (P<0 .0 1或 0 .0 5 ) ,切割海马伞侧隔区 Ch AT和 NOS阳性神经元数均较对照组多 (P均 <0 .0 1) ,其胞体和纤维萎缩程度也较对照组轻。结论 :以党参、黄芪、何首乌、山萸肉等为主方的中医药对切割海马伞大鼠的学习。Objective: Studying the effects of the traditional Chinese medicines consisting of Codonopsis pi'osula Nannf, Astragalus membranaceus Bge,Polygonum multiflorum Thunb,Cornus officinalis sieb.et Zncc, etc. on the learning and memory and the prevention of the degeneration of septal area cholinergic neurons of the rats following fimbrial transection.Methods: The experiments were divided into exprimental and control groups. The traditional Chinese medicines and the water were respectively forced down two groups' rats before and after the fimbrial transection. Step-through passive avoidance and active avoidance tests were respectively carried at the third and forth week after the operation of fimbrial transection. Lastly, the animals were executed. Septal area brain sections were stained with ChAT immunohistochemistry and NOS histochemistry. The numbers of ChAT and NOS positive neurons on the side of fimbrial trasection were counted in the medial septum and vertical limb of diagonal band of Broca and the changes of neuronal bodies and fibers were observed under microscopye.Results: The acquisition frequency and retention time of Step-through passive avoidance test and the positive rates of active and total avoidance reaction of active avoidance test in the experimental group were all superior to those of control group (P<0.01 or 0.05). The numbers of ChAT and NOS positive neurons on the side of fimbrial transection in the experimental group were more than those of the control group (P<0.01). The atrophy of neuronal bodies and fibers in the experimental group was lighter than that of the control group.Conclusion: The traditional Chinese medicines consisting of Codonopsis pi'osula Nannf, Astragalus membranaceus Bge,Polygonum multiflorum Thunb,Cornus officinalis sieb.et Zncc, etc. may improve the learning and memory of the rats following fimbrial transection and protect some septal area cholinergic neurons of these rats from the neuronal death.
