摘 要:目的 :探讨去甲肾上腺素 (NA)的不同浓度、不同作用时间及肾上腺素能受体对培养 T细胞功能的影响 ,以了解神经递质 NA对细胞免疫功能的调节作用及其机制。方法 :大鼠脾细胞用刀豆素 A(Con A)刺激并培养 72 h,在刺激的开始加入 10 - 1 0~ 10 - 5 mol/L NA,或在刺激的不同时间加入 NA,或者再加入 α和 β肾上腺素能受体拮抗剂 ,然后应用噻唑蓝 (MTT)比色分析法检测 T细胞的转化反应。结果 :10 - 8~ 10 - 5 m ol/LNA均可显著抑制培养 T细胞的功能 ,其中 10 - 7m ol/L NA的抑制作用最强。 NA与先被 Con A刺激 1h后的T细胞作用 72 h左右 ,或者 NA与脾细胞作用 1h后被洗去 ,T细胞功能仍受到明显抑制 ,但 NA不能抑制先被 Con A刺激 6 h后的 T细胞转化。α肾上腺素能受体拮抗剂酚妥拉明和 β肾上腺素能受体拮抗剂心得安均可完全阻断 NA抑制 T细胞功能的作用。结论 :NA具有非浓度依赖性抑制培养 T细胞功能的作用 ,此作用可能发生于 T细胞被激活前或者 T细胞被激活早期 ,并可能通过α和β肾上腺素能受体介导。Objective: The effect of noradrenaline (NA) at various concentrations and different action times as well as adrenergic receptors on T lymphocyte function in vitro was investigated in order to know the regulatory role and mechanism of neurotransmitter NA on cellular immunity. Methods: Rat splenocytes were stimulated by concanavalin A (Con A) and incubated for 72 hours. NA at 10 -10 to 10 -5 mol/L concentrations or different time after Con A stimulation was added to splenocyte suspension, or adrenergic receptor antagonist was added to splenocyte suspension containing NA. MTT colorimetric assay was used to determine the conversion of rat splenocytes. Results: NA, in the range of 10 -8 to 10 -5 mol/L, significantly attenuated the conversion of T lymphocytes and the strongest inhibition emerged at 10 -7 mol/L NA. T lymphocyte conversion was still diminished either when NA acted with T cells having stimulated by Con A for an hour or when NA was washed off after one-hour incubation with T cells. However, the T cell conversion did not change when NA acted with T cells having incubated with Con A for 6 hours. The NA suppression of T cell conversion could be blocked either by α-adrenergic antagonist phentolamine or by β-adrenergic antagonist propranolol. Conclusion: It is suggested that NA exert a concentration-independent inhibitory effect on the T lymphocyte function in vitro, which may be related to the early events involved in the initiation of the T cell conversion and be mediated by either alpha- or beta- adrenergic receptors.
