摘 要:目的:研究二甲双胍(MF)对去卵巢模型大鼠血清中瘦素含量的影响及其机制。方法:取雌性大鼠行卵巢摘除术建立去卵巢模型,随机分为模型组和MF低、高剂量[135、270mg(/kg·d)]组,每组10只,另选健康雌性大鼠10只行假手术作为假手术组。模型组和假手术组大鼠灌胃0.9%氯化钠溶液5ml(/kg·d),MF低、高剂量组大鼠灌胃相应药物,连续给药42d。放射免疫法测定各组大鼠血清中瘦素含量并称体质量,逆转录-聚合酶链式反应技术检测各组大鼠脂肪中细胞因子信号传导抑制因子3(SOCS-3)mRNA的表达。结果:与假手术组比较,模型组大鼠血清中瘦素含量、体质量和脂肪内SOCS-3mRNA表达均明显增加(P<0.05或P<0.01);与模型组比较,MF高剂量组大鼠血清中瘦素含量、体质量和脂肪内SOCS-3mRNA表达均明显降低(P<0.05),MF低剂量组大鼠上述指标差异无统计学意义(P>0.05)。结论:高剂量MF可明显降低去卵巢模型大鼠血清中瘦素含量,其机制可能与显著下调脂肪内SOCS-3mRNA表达有关。OBJECTIVE: To study the effects of metformin (MF) and its mechanism. METHODS: Female rats underwent ovariosteresis on the serum content of leptin in ovariectormized model rats surgery to establish ovariectomized model. Model rats were randomly divided into model group, MF low-dose and high-dose groups [135, 270 mg/(kg.d)] with 10 rats in each group; other 10 healthy female rats underwent sham operation were sham group. Model group and sham group were given 0.9% Sodium chlo- ride 5 ml/(kg-d) intragastrically, and MF low-dose and high-dose groups were given relevant drugs intragastrically for consecutive 42 days. The serum content of leptin was measured with radioimmunoassay and body weight of rats were measured; the expression of SOCS-3 mRNA in lipid was measured with RT-PCR. RESULTS: Compared with sham group, the serum levels of leptin, body weight and the expression of adipose SOCS-3 mRNA increased significantly in model group (P〈0.05 or P〈0.01). Compared with model group, the serum levels of leptin, body weight and the expression of adipose SOCS-3 mRNA decreased significantly in MF high-dose group (P〈0.05) ; but above index had no statistical significance in MF low-dose group (P〉0.05). CONCLUSIONS: The high-dose of MF can significantly decrease the serum levels of leptin in ovariectormized model rats, and it may be related to the down-regulation of the expression of SOCS-3 mRNA.
