摘  要:目的 :对神经生长因子 (NGF)及其结合肌基膜管 (MBL)修复脊髓横断性损伤进行组织学评价。方法 :以雌性家犬为实验对象 ,手术将其脊髓横断 ,随机分为 3组 :( 1) 7只行MBL移植结合注射NGF(A组 ) ;( 2 ) 6只单纯注射NGF组 (B组 ) ;( 3) 4只为对照组。 6个月后用免疫组化对神经轴突和胶质细胞网格框架结构进行特异染色 ,并用图像分析方法对脊髓横断处的远、近端横截面神经纤维进行定量研究。结果 :各组间远端神经纤维数量均有显著性差异 ;MBL移植物中有神经纤维通过 ,对照组断端为坏死空腔。电镜证实横断远端存在新生神经纤维。结论 :MBL移植结合NGF修复脊髓横断性损伤 ,神经轴突能越过移植区到达远端 ;Objective:To observe the histological changes of transplanting autologous muscle basal lamina(MBL)and using nerve growth factor(NGF) to repair spinal cord transection in the adult dog.Methed:Laminectomy was done in 17 adult female dogs over T13 and L1.After complete transection of the spinal cord at T13 level.The animals were divided into three groups.Group A:7 dogs,a gap of 1cm was done longitudinally on the spinal cord at the lesion site.Autologous MBL grafts made from paraspinal muscle fibers were implanted to the spinal cord gaps.NGF was administered at the lesion site via indwelling,implaned,osmotic minipumps.Purified NGF was supplied at dose of 20μg once a week for 4 weeks.Group B:6 dogs,NGF was administered at the lesion site via indwelling,implaned,osmotic minipumps.Purified NGF was supplied at the same dose.Group C:4 dogs,served as controls and each animal received a complete transection on the spinal cord.Neurological function was observed for 6 month and the animals were sacrificed.The grafted materials and treated spinal cord were collected for histological examination.All of the slices were processed on immunohistocytochemical staining,using monoclonal antibody to neurofilament(NF).The charactoristics of regeneration was observed histologically by immunohistocytochemistry methods and electron microscope.All of the immunostaining pictures were analyzed by the method of morphometric analysis which gave out the data of the axons counting.Result:The results showed significantly better microscopic and electron microscopic appearances in group A with implantation of both MBL and NGF.Nerve fibers were identified in the graft by immunohistocytochemical staining for NF.Degeneration and necrosis were found in group C.Little degeneration and necrosis occurred in group B and group A.Conclusion:These data showed that MBL can provide a suitable environment for axonal regeneration and NGF is able to enhance the regeneration capacity of central nevous system.Transplant of autologous MBL and use of NGF to repair spi
