摘  要:目的:探讨益肾康对单侧输尿管梗阻(UUO)大鼠肾组织肝细胞生长因子(HGF)、单核细胞趋化蛋白(MCP-1)及其mRNA表达的影响。方法:50只雄性SD大鼠随机分为假手术组、模型组、益肾康大剂量组、益肾康小剂量组、福辛普利组。每组10只,除假手术组外均建立UUO模型,术后21 d腹主动脉采血,检测尿素氮(BUN)、血肌酐(Scr)、谷丙转氨酶(ALT)、谷草转氨酶(AST)的变化;用免疫组化法检测HGF、MCP-1的表达部位及含量,逆转录-多聚酶链反应(RT-PCR)检测HGF和MCP-1mRNA表达水平。结果:UUO组大鼠肾间质MCP-1及mRNA表达显著高于假手术组(P<0.01);药物干预组MCP-1及mRNA表达明显低于模型组(P<0.01);益肾康大剂量组低于小剂量组(P<0.01),但与福辛普利组相似(P>0.05);药物干预组HGF及mRNA含量显著升高(P<0.01),其中益肾康大剂量组的表达高于小剂量组(P<0.01),与福辛普利组相似(P>0.05)。药物干预组BUN水平下降,Scr、ALT、AST变化不明显。结论:益肾康可通过上调UUO大鼠肾组织HGF及其mRNA表达,抑制UUO大鼠肾组织MCP-1及其mRNA表达而实现抗肾小管间质纤维化的作用,益肾康大剂量组与福辛普利组作用相似,且没有明显的肝肾毒性。Objective:To observe the effects of yisenco asiatica on the expression of HGF,MCP-1 and it's mRNA in the tubulointerstitial of unilateral ureteral obstruction (uUO) rat. Methods: 50 Sprague-Dalwley (SD) male rats were randomly divided into sham-operated group, model group, high centella asiatica group,low centella asiatica group, fosinopril group. 10 of each group, all of the rats were set up UUO model except the sham-operated group. 21 days later, blood sample was collected from each mouse to detect the change of BUN,Scr,ALT,AST; the same time the obstructed kidney was taken, part of them was used to examine the expression of HGF,MCP-1 by immunohistochemical method, the other part was transferred to the ultra-low temperature refrigerator to detect the expressions of HGF,MCP-lmRNA by the method of RT-PCR. Results The level of MCP-1 and MCP-lmRNA in the UUO group increased remarkably compared with that in the sham-operated group (P 〈0. 01 ). Drug intervention group was obviously low compared with model group ( P 〈 0.01 ), high yisenco asiatica group was below with it' s low group (P 〈 0.01 ) ;but it was similar with fosinopril group (P 〉 0.05 ) ; The expression of HGF had only a litter in the blank group , The level of HGF and it' s mRNA in the drug group increased remarkably compared with that in the sham-operated group ( P 〈 0.01 ). And the high yisenco asiatica group was higher compared with it' s low group (P 〈 0. 05) ;but the same with fosinopri group ( P 〉 0.05) ; the lever of BUN in the drug group was discreased, and the Scr,AST,ALT had no change. Conclusion: Yisenco asiatica can inhibit renal intersititiun fibrosis caused by UUO rat by reducing MCP-1 protein and MCP-1mRNA expression,and inhibit renal intersititiun fibrosis caused by UUO rat by increasing HGF protein and HGF mRNA expression, the high dose is similar with fosinopril, and has no harm for the kidney and liver.
