摘  要:目的:观察姜黄素对转化生长因子β1(TGF-β1)诱导的大鼠肾小球系膜细胞增殖及TGF-β1/Smads信号通路的影响。方法:采用TGF-β1诱导肾小球系膜细胞增殖和分泌胶原,实验分为空白对照组、单纯TGF-β1刺激组、姜黄素组3组。MTT法检测药物对肾小球系膜细胞增殖的抑制率,ELISA法检测肾小球系膜细胞Ⅰ型胶原的表达,Western blot法检测肾小球系膜细胞pSmad2的表达。结果:姜黄素能够显著抑制TGF-β1诱导的肾小球系膜细胞增殖(P<0.01)及Ⅰ型胶原分泌,并明显下调pSmad2蛋白表达。结论:姜黄素能部分阻断TGF-β/Smads信号传导通路的激活,抑制Ⅰ型胶原分泌及肾小球系膜细胞增殖。Objective :To observe the effect of curcumin on TGF-β1 (transforming growth facto -β1, TGF-β1 )induced proliferation and Smads pathway in rat GMC (glomerular mesangial cell, GMC). Methods:The experiment was divided into 3 groups:blank,TGFβ1 control,curcumin groups. MTT method was adopted to examine the proliferation and inhibition rate of GMC. Enzyme-linked immunosorbent assay was used to determine the production of type I collagen in cultured GMC. The production of pSmad2 in GMC was detected by western blot. Results: Curcumin significantly inhibited the proliferation of GMC(P 〈 0. 01 ) and also inhibited the production of type I collagen, downregulated the expression pSmad2. Conclusion: Curcumin can partially block the activation o f pSmad signaling pathway in GMC, inhibit the excretion of collagen I and proliferation of GMC.
