清热活血方在类风湿关节炎相关动物实验和体外研究中对白介素17的影响

摘　　要：目的:探讨清热活血方(QRHX)在类风湿关节炎相关动物实验和体外研究中对白细胞介素17(IL-17)的影响。方法:(1)动物实验:SD大鼠46只,随机分为正常组、模型组、模型QRHX组、模型甲氨蝶呤组,规律灌胃给药6周后观察大鼠血清IL-17的表达;(2)体外研究:不同体积分数的QRHX含药血清与类风湿关节炎患者成纤维样滑膜细胞(RA-HFLS)共孵育,ELISA法检测培养液上清中IL-17的表达。结果:QRHX给药组大鼠血清IL-17的含量较模型组明显降低(P<0.01),QRHX含药血清可显著抑制共培养系统中IL-17的分泌,并呈现一定的浓度依赖性(P<0.01)。结论:清热活血方抑制RA骨破坏的护骨作用可能是通过降低IL-17表达而发挥作用。Objective： To investigate the effects of Qing-re-huo-xue decoction （QRHX, the Decoction with the function of clearing heat ＆ promoting blood flow） on the level of interleukin-17 （IL-17） in CIA rats and in vitro studies. Method： ① Animal experiments ： 46 SD rats were randomly divided into normal group, model group, groups of QRHX, and group of methotrexate, medicine filling for six weeks; then tested the serum IL-17 level. ② In vitro studies： QRHX containing serum with different concentrations were co-cultured with human fibroblast-like synoviocytes of rheumatoid arthritis （ RA- HFLS） and tested IL-17 level in culture supernatant with ELISA. Results： ① QRHX suppresses serum IL-17 levels in comparison with the model group （P 〈 0. 01 ） ; ② QRHX containing serum inhibit IL-17 secretion in co-culture system, and presents some concentration-dependent＇.manner （P 〈0. 01 ）. Conclusion： Qing-re-huo-xue decoction slows down RA bone destruction may take effects through the way of reducing the expression of IL-17.
