摘 要:目的:通过从脐带中分离和培养脐带间充质干细胞(MSCs),探讨其对再生障碍性贫血(再障)小鼠骨髓造血的影响。方法:原代贴壁法培养人脐带MSCs,将小鼠分为模型组、对照组、MSCs输注组进行实验。经静脉输注免疫介导方法建立再障模型小鼠,分别于3、7、14、21、28 d观察其外周血象指标变化和骨髓组织形态;ELISA检测造血负调控因子(IFN-γ)的水平,计数骨髓造血干细胞集落生成单位。结果:原代培养中脐带组织块直接贴壁后8~10 d,边缘爬出长梭形MSCs,随传代培养呈平行、漩涡状生长;再障小鼠经MSCs输注治疗后外周血象明显增高,IFN-γ水平下降(P<0.01);治疗组骨髓红细胞集落形成单位与粒-巨噬细胞集落形成单位均较模型组明显增高(P<0.01),成纤维细胞集落形成单位计数差异无统计学意义(P>0.05)。结论:人脐带来源的MSCs对再障模型小鼠骨髓可发挥造血支持作用。Objective: To investigate the effects of umbilical cord mesenchymal stem cells (MSCs)on hemopoietic function of bone marrow in mice with aplastic anemia (AA). Methods:Umbilical cord MSCs were primarily cultured with adherent method. All mice were divided into the model group, control group and MSCs infusion group. The aplastic anemia model mice established by immune- mediated means were infused with MSCs through caudalis vena. The peripheral hemogram and bone marrow morphology were observed at 3,7,14,21 and 28 days. The level of hematopoiesis negative regulatory factor(IFN-γ) was detected by ELISA. The colony-forming unit counts of bone marrow hemopoietic progenitor cells were measured. Results:The spindle-shaped MSCs extended from the tissue edge at 8 - 10 days after primary adherent culture of umbilical cord, and those were parallelly or whirlpool-shape growth with the cultivation. The increasing peripheral hemogram and decreasing IFN-γ levels of AA mice treated with MSCs were significant (P 〈 0.01 ). The colony forming units-erythroid and colony forming units-granulocyte macrophage of the treatment group were significantly higher than those in the control group (P 〈 0.01 ) , but there was no significant difference in the colony forming units-fibroblast count between the two groups (P 〉 0.05). Conclusions: MSCs derived from human umbilical cord can play some poles on hemopoietic function of the bone marrow of AA mice.
