摘  要:鳞柄白鹅膏为外生菌根菌。为了确定试验菌株是否为鳞柄白鹅膏(Amanita virosa),对其菌丝体进行了ITS序列测定和GenBank基因库比对,并通过生长速率法研究其室内培养条件。结果显示,试验菌株的ITS序列长度为516bp,在GenBank核酸序列数据库中比对,试验菌株与鳞柄白鹅膏的相似率为95%,可以确定此菌株为鳞柄白鹅膏,GenBank登录号为EF493032。pH值对鳞柄白鹅膏培养菌丝生长的影响根据不同的培养基而不同,其中在含马铃薯的培养基中,pH值对菌丝生长的影响很小,pH值在5~10时均生长良好,可以作为后续试验的主要培养基。供试最适碳源为甘露醇,最适氮源为酵母膏,最适生长温度为25℃。Amanita virosa is ectomycorrhizal fungi.ITS sequence determination and blast searching in Genbank databases were carried out for the test strain's identity to determine weather the test strain was Amanita virosa.The optimum nutritional and culture conditions were tested by studying the effects on mycelial growth of the test strain,which provided the materials for the further study.The mycelium of the tested strain was ITS sequenced,revealing the length of rDNA ITS sequence of 516 bp.After sequence alignment in GenBank database,the similarity of tested strain and A.virosa was 95% and the tested strain was identified as A.virosa.The GenBank accession number was EF493032.The effect of pH on the mycelial growth was different with the different media;in the medium containing potato,when pH was 5-10,the mycelial growth was well.So the three kinds of media containing potato could be used as the main medium for next researches.The optimum culture temperature was 25 ℃,the optimum carbon source was mannitol and nitrogen source was yeast extract.
