摘 要:目的 研究高碘对豚鼠脑和甲状腺细胞凋亡的影响。方法 用高碘水喂养豚鼠复制出高碘甲状腺肿模型 ,采用光镜及透射电镜形态学观察、DNA凝胶电泳及流式细胞术 ,分两期观察不同浓度高碘水对大脑皮质、海马和甲状腺细胞凋亡的影响。结果 1高碘可以诱发豚鼠大脑皮质、海马和甲状腺细胞凋亡 ,呈现凋亡形态学及生化特征性改变。2高碘所诱发的大脑皮质、海马和甲状腺组织的细胞凋亡率高低与摄碘量和碘作用时间的长短有关。结论 1高碘可以诱发大脑皮质、海马和甲状腺细胞凋亡。2高碘通过诱发大脑皮质、海马和甲状腺细胞凋亡的发生而对甲状腺及中枢神经系统造成损伤 。Objective To investigate the effect of excessive iodine on apoptosis in brain and thyroid.Methods An animal model of excessive iodine goiter was reproduced in Guinea pig by feeding water containing high level of iodine for 90 and 180 days,respectively.Using techniques of flow cytometry,DNA agarose gel electrophoresis and transmission electron microscope (TEM),we investigated the effect of excessive iodine on apoptosis in cortex,hippocampus and thyroid.Results The results showed that:①Excessive iodine may induce apoptosis in thyroid and brain with the characteristic changes of morphology and biochemistry.②The apoptosis rate in cortex,hippocampus and thyroid has certain relation to the content of iodine and exposure time.Conclusions ①Excessive iodine may induce apoptosis in thyroid and brain.②By inducing the apoptosis in brain and thyroid,excessive iodine may not only damage thyroid but also damage central nerve system,which lead to impediment of its function.
