摘 要:目的 :优化穿龙薯蓣总皂甙水解条件 ,以提高薯蓣皂甙元的含量与得率。方法 :以薯蓣皂甙元的含量为指标 ,应用L9(34 )正交试验设计筛选穿龙薯蓣总皂甙水解条件。结果 :影响水解的主次因素为 :D >A >B >C(A为硫酸浓度 ;B为稀硫酸用量 ;C为回流时间 ;D为与水不相溶的溶剂用量 )。最佳水解条件为 :各加 2 %硫酸和石油醚 (6 0~ 90°C) 10 0ml,回流 4h。结论 :采用优化后的水解条件明显提高薯蓣皂甙元的含量 。Objective: To optimize hydrolysis conditions of total saponins in Dioscorea nipponica Makino so as to raise the content and yield of diosgenin. Methods: The optimum hydrolysis conditions were investigated by the orthogonal design and in diosgenin content as a marker. Results: The factors influencing hydrolysis extraction efficiency were as follows: D>A>B>C (A:H 2SO 4 concentration. B: the amount of dilute H 2SO 4. C: refluxing time. D: the amount of solvent which doesn't dissolve in water). The optimum hydrolysis condition obtained was :refluxing for 4 hours with 100ml of 2% dilute H 2SO 4 solution plus 100ml of pertroleum ether (60~90 °C ). Conclusion: The content of diosgenin under the optimum hydrolysis condition is raised with a good reproducibility.
