类风湿关节炎患者外周血髓源性抑制细胞的检测及与Th1/Th2细胞的相关性分析

摘　　要：目的:检测类风湿关节炎(rheumatoid arthritis,RA)患者外周血中髓源性抑制细胞(myeloid-derived suppressorcells,MDSCs)的水平,分析其与Th1/Th2细胞的相关性。方法:选取确诊的RA患者36例,根据临床资料计算DAS28评分,将其分为活动组和稳定组;另选取10例健康者作为对照组。3组MDSCs、Th1及Th2细胞的比例用流式细胞术检测并进行比较。分析MDSCs与Th1/Th2细胞的相关性。结果:活动组MDSCs的比例明显高于稳定组和对照组(P均<0.05);稳定组MDSCS的比例与对照组比较,差异无统计学意义(P>0.05)。经Spearman相关性分析,MDSCs与Th1细胞呈负相关(P<0.05);而与Th2细胞无相关性(P>0.05)。结论:MDSCs在RA活动期显著增高,且与CD4+T细胞亚群关系密切,可作为辅助评估RA活动状态的指标。Objective： To investigate the frequency of myeloid-derived suppressor cells（MDSCs） in the peripheral blood from patients with rheumatoid arthritis（RA） and assess the correlations between MDSCs and Th1/Th2 cells.Methods： A total of 36 RA patients were enrolled in the study.Disease activity was evaluated by calculating the 28-joint Disease Activity Score（DAS28） based on clinical information.RA patients were divided into active group and inactive group according to the DAS28 score.The frequency of MDSCs and Th1/Th2 cells in peripheral blood from RA patients and healthy controls were determined by flow cytometry.Results： The frequency of MDSCs increased significantly in active group compared with both inactive group and control group（P＆lt;0.05）.However,there was no significant difference in the frequency of MDSCs between inactive group and control group（P＆gt;0.05）.The frequency of MDSCs was negatively correlated with the frequency of Th1 cells in peripheral blood,but not correlated with the frequency of Th2 cells by Spearman′s test.Conclusion： The frequency of MDSCs in peripheral blood from RA patients is highly prevalent and closely related to helper T cells,which may become a valuable marker for the evaluation of RA.
