肺积方调节CD4+CD25+Treg细胞及T淋巴细胞增殖与抗Lewis肺癌移植鼠肿瘤生长作用研究

摘　　要：目的:观察肺积方联合化疗对Lewis肺癌移植小鼠肿瘤生长情况及脾脏CD4+CD25+Treg细胞、胸腺T淋巴细胞增殖变化的影响,探讨肺积方联合化疗抗肺癌免疫逃逸的作用机制。方法:采用随机对照研究方法,将64只Lewis肺癌移植小鼠随机分为对照组、肺积方组、综合组(肺积方联合顺铂)、顺铂(DDP)组,每组16只,并分别以生理盐水、肺积方、肺积方联合DDP、DDP药物干预,观察小鼠瘤体变化,流式细胞术检测小鼠脾脏CD4+CD25+Treg细胞含量,MTT法检测小鼠胸腺和脾脏T淋巴细胞增殖情况。结果:DDP组、肺积方组、综合组小鼠瘤体、瘤重均小于对照组(P<0.05),综合组瘤体、瘤重均小于DDP组和肺积方组(P<0.01);第10、21天DDP组、肺积方组、综合组小鼠胸腺、脾脏T淋巴细胞增殖指数与对照组比较,差异有统计学意义(P<0.05),其中肺积方组、综合组T淋巴细胞增殖指数增高,而DDP组T淋巴细胞增殖指数降低;DDP组干预后T淋巴细胞增殖指数明显低于干预前(P<0.05);而肺积方组、综合组第21天与第10天T淋巴细胞增殖指数组内比较无明显变化(P>0.05)。第10、21天DDP组、肺积方组、综合组荷瘤小鼠胸腺CD4+CD25+Treg细胞比例均较对照组降低(P<0.05);DDP组第21天胸腺CD4+CD25+Treg细胞比例较第10天明显升高(P<0.05);综合组脾脏CD4+CD25+Treg细胞比例较低,与肺积方组、DDP组比较差异有统计学意义(P<0.05)。结论:肺积方联合化疗具有抗Lewis肺癌移植小鼠肿瘤生长的作用,该作用可能与其下调CD4+CD25+Treg细胞含量相关。Objective：To investigate the effect of Feijifang combined with chemotherapy on tumor growth and the levels of CD4＋CD25＋ regulatory T cells（Treg） and T lymphocytes in mice with transplanted Lewis lung cancer and the underlying mechanism of Feijifang modulating immune evasion. Methods：Sixty-four mice with transplanted Lewis lung cancer were randomly divided into control group,Feiji group,comprehensive treatment group（Feijifang combined with diamminedichloroplatinum）,and diamminedichloroplatinum （DDP） group,16 mice in each. The proliferation of T lymphocytes in the spleen and thymus was detected by MTF method and the percentage of CD4＋CD25＋Treg was assessed by flow cytometry. The volume and weight of tumors were recorded and evaluated. Results：The volume and weight of tumors in the DDP,Feiji,and comprehensive treatment groups were less than those in the control group （P〈0.05）. The volume and weight of tumors in the comprehensive treatment group were less than those in the Feiji and DDP groups（P〈0.01）;On day 10 and 21 after intervention, a significant difference in the level of T lymphocytes was noted between the control group and the DDP, Feiji, and comprehensive treat-ment groups（P〈O.05）;the level of T lymphocytes increased in the Feiji and comprehensive treatment groups but de- creased in the DDP group （P〈0.05）;and no significant difference was found between the level on day 10 and the level on day 21 in the Feiji and comprehensive treatment groups （P〉0.05）. On day 10 and 21,the percentage of CD4＋CD25＋Treg was lower in the DDP,Feiji,and comprehensive treatment groups as compared with that in the control group （P〈O.05）;in the DDP group,the percentage of CD4＋CD25＋Treg increased after intervention as com- pared with that before intervention（P〈0.05）;and the percentage of CD4＋CD25＋Treg was lower in the comprehensive treatment group than that in the DDP and Feiji groups（P〈0.05）.
