摘 要:目的使用乙二胺四乙酸二钠(ethylene diamine tetraacetic acid disodium salt,EDTA-Na2)体外脱钙法制备成年猪椎体骨质疏松模型,探讨一种快速、有效制备离体椎体骨质疏松生物力学模型的方法。方法选取成年猪胸腰段椎体36个,甲醛浸泡固定后随机分成四组,每组9个椎体,采取4种处理方法:A组为对照组,不做处理,B、C、D组分别以0.595 mmol/L的EDTA-Na2溶液脱钙10 d、15 d、20 d。后对其进行大体、直接数字化X射线(digital radiogrx-phy,DR)观察,并行骨矿密度(bone mineral density,BMD)测定,再行垂直压缩试验检测椎体强度及刚度,观察其随时间延长而发生的变化。结果大体标本观察:随着脱钙时间的延长,椎体椎弓根处钉孔依次扩大,脱钙液浸润范围依次增加;DR显示:脱钙组椎体骨密度显著降低,骨量减少;随着脱钙时间的延长、椎体BMD、压缩强度和刚度呈依次降低的趋势,组间比较有统计学意义(P<0.01)。结论应用EDTA-Na2脱钙法可在较短时间内建立用于生物力学研究的骨质疏松性椎体模型。Objective To use EDTA-Na: prepared in vitro demineralized aduh pig vertebral osteoporosis model, to explore a rapid,effective method for preparati#n :of isolated vertebral osteoporosis biomechanical model. Methods 36 adult porcine thoracolumbar vertebral, formaldehyde after soaking were randomly divided into 4 groups, with 9 vertebral bodies in each group. There were 4 treatment methods:Group B was treated with 0. 595 mmol/L EDTA-Naz solution for 10 days. Grop C was treated with the same solution for 15 days, And grop D for 20 days. After the general and DR observation, bone mineral density (BMD) were measured,and vertical compression test was used to detect vertebral body strength and stiffness. Results The gross observation showed that with the decalcification time prolonged ,vertebral pedicle nail holes enlarged ,decalcified fluid erosion increased;DR showed than in demineralized group, vertebral bone mineral density was significantly decreased, vertebral BMD, compression strength and stiffness of the decalcification time showed a decrease trend. There was statistical significance between groups ( P 〈 0.01 ). Conclusion The application of EDTA-Na2 decalcification method can be established for osteopo- rotic vertebral model biomechanies research in a relatively short period of time.
