奥氮平抑制线索诱导的大鼠海洛因觅药行为

摘　　要：目的獉獉:研究奥氮平对线索诱导或海洛因引燃的大鼠觅药行为的影响。方法獉獉:大鼠用固定比率程序进行海洛因自身给药训练14 d,然后环境消退10 d,随后在线索诱导的或海洛因引燃的觅药行为测试前给予不同剂量的奥氮平,观察奥氮平对大鼠觅药行为的影响。结果獉獉:0.3 mg.kg-1的奥氮平能显著抑制线索诱导的大鼠觅药行为(P<0.01),但对海洛因引燃的大鼠觅药行为无抑制作用,且0.3 mg.kg-1的奥氮平对大鼠的蔗糖自身摄取行为和自发活动度都无明显的抑制作用。结论獉獉:低剂量的奥氮平能够降低药物相关的条件性线索诱导的海洛因复吸行为,奥氮平有可能成为治疗海洛因成瘾的新的辅助药物。Objective：The aim of the present study was to investigate the effects of pretreatment with olanzapine on heroin - seeking behaviour induced by cues or heroin priming. Meth_o~： Rats were trained to self - administer heroin under a fixed ratio 1 schedule for 2 weeks and nose - poke responding was extinguished for 10 d,after which the rats were injected with vehicle or olanzapine 30 min prior to the reinstatement test. The reinstatement of drug seeking induced by conditioned cues or heroin priming was observed for 2 h. Results ： Olanzapine at the dose of 0. 3 mg ~ kg-1 significantly inhibited reinstatement of conditioned cue - induced heroin seeking （ P 〈 0. 01 ）, but did not attenuate reinstatement induced by heroin priming. Olanzapine at this dose failed to influence sucrose self - adminstration and locomotion activity. Conclusion： Acute treatment with low - dose olanzapine inhibits conditioned cue - induced heroin seekin~ and olaza^ine may be an adjunctive dru~ for the treatment of heroin addiction.
