摘  要:采用原代培养的兔关节软骨细胞,检测硫酸氨基葡萄糖氯化钾对IL-1β诱导造成的炎性反应的保护作用。加入0.2,2,20μmol/L硫酸氨基葡萄糖氯化钾及10 ng/mL IL-1β48 h后,测定蛋白聚糖酶(Aggrecanase-1,-2)和诱导型一氧化氮合酶(iNOS)mRNA表达量的变化。结果表明,硫酸氨基葡萄糖氯化钾对于兔软骨细胞无明显增殖和细胞毒作用。对于IL-1β诱导的软骨细胞,硫酸氨基葡萄糖氯化钾可抑制其聚蛋白聚糖酶Aggrecanase mRNA表达量,并可抑制NO释放以及iNOS mRNA表达,与IL-1β模型组相比有极显著差异。硫酸氨基葡萄糖氯化钾保护骨性关节炎的机制与可显著抑制由白介素类炎性因子导致的炎性反应相关。Primary cultured rabbit chondrocytes were used to study the protective effect of glucosamine sulfate potassium chloride on IL-1β induced inflammatory reaction.After being cultured with 0.2,2,20 μmol / L of glucosamine sulfate potassium chloride and 10 ng / mL of IL-1β for 48 hours,Aggrecanase-1,-2 and iNOS were tested in RNA level in different groups.The results showed that 0.2,2,20 μmol / L of glucosamine sulfate potassium chloride had no cytotoxic on rabbit chondrocytes.After 2,20 μmol / L glucosamine sulfate potassium chloride or 40 μmol / L glucosamine hydrochloride combined with 10 ng / mL IL-1β treated,the RNA level of aggrecanase-1,-2 and iNOS had significant differences from the IL-1β induced control group.The release of NO was also tested to have significant differences from the IL-1β induced control group.The mechanism may be related to IL-1β inducement of inflammatory reaction.
