摘 要:促分裂原活化蛋白激酶(MAPK)在介导植物应答各种发育和环境信号过程中起重要作用。在富集低磷逆境特异表达基因的小麦(‘石新828’)根系差减cDNA文库中,获得了1个与短柄草MAPK基因(MAPK7-1)高度同源的表达序列标签(EST),进一步在小麦中鉴定及克隆了与该EST对应的MAPK基因(TaMAPK7-1)。TaMAPK7-1的cDNA全长为1 611bp,编码369个氨基酸残基,编码蛋白含有1个TEY基序(aa192至aa194)。表明TaMAPK7-1具有应答特定上游MAPKK激酶信号的潜在功能。小麦根、叶中TaMAPK7-1在低磷和高盐逆境下的表达较处理前均明显增强,在低氮和干旱逆境下的表达与处理前对照相比没有改变。研究表明,TaMAPK7-1通过对低磷和高盐逆境产生明显应答,参与小麦上述逆境信号转导过程,在小麦应答和抵御上述逆境中可能发挥着重要作用。Mitogen-activated protein kinases (MAPKs) play an important role in mediating the transduction of signals that are initiated by development cues and environmental factors in plants. In this study, an expressed sequence tag (EST) sharing high similarity to MAPK7-1, a MAPK gene characterized in Brachypodium, was identified in a wheat ('Shixin 828~) root subtractive suppression hybridization cDNA library. The gene corresponding to this EST was further obtained and denoted as TaMAPK7-1. The full-length cDNA of TaMAPK7-1 is 1 611 bp long, encoding a 369-aa polypeptide. A conserved motif acting as the symbol of MAPK re- ferred to TEY is located at TaMAPKT-1, suggesting that this wheat gene has a potential function in responding distinct upstream signalings. Under low-Pi and high-salinity stresses, the TaMAPKT-1expression levels in the wheat roots and leaves were both up-regulated compared with those before the treatments. No variations on the TaMAPK7-1 expression levels in the roots and leaves were found af- ter the treatments of low-N and drought compared with those before the treatments. Therefore, TaMAPK7-1 is responding to stresses of low-Pi and high-salinity, and is involved in mediating the transduction of signals that are initiated by aforementioned stresses. TaMAPKT-1 has a potential role in improving the plant tolerance to low-Pi and high-salinity in wheat.
