摘  要:为研制H9亚型禽流感和新城疫的重组二联活疫苗,从农业部畜禽传染病重点开放实验室2012年分离的H9亚型禽流感病毒中选取一株具有代表性的流行株A/Chicken/Jiangsu/WJ57/2012(H9N2)作为HA基因的供体。在新城疫弱毒LX株反向遗传操作平台的基础上,构建了表达该株H9亚型禽流感病毒HA基因的重组新城疫病毒基因组全长cDNA克隆pLX-H9HA。拯救出的重组嵌合体病毒rLX/H9HA经间接免疫荧光鉴定,并对其生物学特性、安全性和免疫原性进行测定。结果表明:获救的重组病毒呈典型弱毒特征,同时保持了亲本毒株的高滴度生长特性和遗传稳定性,可作为H9亚型禽流感和新城疫二联活疫苗的候选株。To generate a bivalent vaccine against both Newcastle disease (ND)and H9 subtype avian influenza (Al),one representative strain A/ChickerdJiangsu/WJ57/2012 (H9N2)was selected from H9 subtype avian influenza virus (AIV) strains isolated by our lab in 2012 as HA gene donor. On the basis of reverse genetics of NDV strain LX,the recombinant NDV genome cDNA clone pLX-H9HA was constructed, which contained HA gene of this H9 subtype AIV strain. Indirect immunofluorescence assay was used to detect HA protein expression of rescued chimeric virus rLX/HgHA. The biological characteristics were determined and the immunogenicity and safety tests were conducted. The results indicated that the rescued recombinant virus was avirulent,and maintained similar high growing titers and geneticstability as its parent strain did,which suggested that the recombinant virus rLX/H9HA could be served as a live bivalent vaccine candidate against H9 subtype AI and ND.
