摘 要:目的:探讨复方肝毒清的抗肝纤维化机制。方法:以柠檬酸铁胺与RAW264.7细胞共同温育,体外建立铁沉积巨噬细胞模型。将RAW264.7细胞分为空白对照组、铁沉积模型组、中药组、铁沉积模型+中药组共4组,观察复方肝毒清对铁沉积巨噬细胞转化生长因子-β1(TGF-β1)、诱导型一氧化氮合酶(iNOS)、肿瘤坏死因子-α(TNF-α)mRNA表达的影响。结果:铁沉积于巨噬细胞主要使iNOS、TNF-αmRNA表达明显上调,TGF-β1 mRNA表达明显下调,而中药复方肝毒清能使其出现相反基因表达变化。结论:复方肝毒清的抗肝纤维化机制与其降低细胞炎症反应及调节体内铁的代谢有关,而不是通过下调TGF-β1 mRNA的表达来实现。Objective: To investigate the anti-liver fibrosis mechanism of compound Ganduqing decoction. Methods: Iron deposited maerophages model was established by incubating with ferric ammonium citrate (FAC) and macrophages line (RAW264.7 cells) . RAW264. 7 cells were divided into control group, iron deposition model group, Chinese herb group and iron deposition model plus Chinese herb group. Effect of compound Ganduqing decoction on mRNA expression of TGF-β1, iNOS and TNF-ot was investigated. Results: Iron deposition in maerophages mainly up-regulated the mRNA expression of iNOS and TNF-α, and down-regulated the mRNA expression of TGF-β1, where as the opposite gene expression was observed with com- pound Ganduqing decoction therapy. Conclusion: The anti-fibrosis mechanism of compound Ganduqing decoction might be relat- ed to decreasing inflammation response and regulatory action on iron metabolism of maerophages, but not to down-regulation of the mRNA expression of TGF-β1.
