摘 要:目的探讨平喘固本合剂对支气管哮喘(哮喘)小鼠支气管肺泡灌洗液(BALF)中IL-17及呼吸道重塑的影响。方法健康雌性昆明系小鼠40只,随机分为4组:正常对照组、哮喘组、平喘固本合剂干预组和布地奈德雾化干预组,每组各10只。采用卵清清蛋白(OVA)建立小鼠哮喘模型,正常对照组腹腔注射PBS,其余3组腹腔注射OVA混悬液,期间平喘固本合剂干预组每日胃饲平喘固本合剂。在激发阶段,正常对照组雾化吸入PBS,其余3组雾化吸入OVA。布地奈德雾化干预组于激发前雾化吸入布地奈德混悬液。采用ELISA法测定BALF中IL-17水平。免疫组织化学法测定肺组织中基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)水平。结果1.平喘固本合剂干预组和布地奈德雾化干预组BALF中IL-17水平明显低于哮喘组(P均〈0.05);与正常对照组比较,平喘固本合剂干预组BALF中IL-17水平明显升高,差异有统计学意义(P〈0.01)。2.平喘固本合剂干预组和布地奈德雾化干预组呼吸道MMP-9、TIMP-1表达水平和MMP-9/TIMP-1比值明显低于哮喘组,差异均有统计学意义(P均〈0.01);与正常对照组比较,呼吸道MMP.9表达水平和MMP-9/TIMP-1比值明显增高,差异均有统计学意义(P均〈0.01),2个治疗组间比较,差异均无统计学意义(P均〉0.05)。结论平喘固本合剂对哮喘小鼠具有抑制呼吸道炎症和重塑的作用,在一定程度上可替代布地奈德吸人治疗。Objective To study the effect of Pingchuanguben Decoction on IL-17 level of bronchial alveolar lavage fluid (BALF) and airway remolding in mice with asthma. Methods Forty healthy female Kumning mice were randomly divided into 4 groups (10 rats in each group) :normal control group, asthma group, Pingehuanguben Decoction group and Budesonide group. The asthmatic models were established by ovalbumin (OVA) ; the normal control group was treated with phosphate buffer solution (PBS liquid) by celiac injection,during this period Pingchuanguben Decoc- tion was administered to the mice in Pingchuanguben Decoction group each day. In stimulation stage, the normal control group was atomization inhalated PBS liquid, and the rest of the 3 groups were given aerosolized OVA instead. Bude- sonide atomization was administered to mice in Budesonide group before stimulation. The levels of IL-17 in BALF were measured with enzyme-linked immunosorbent assay ( ELISA), and the levels of matrix metalloproteinase-9 ( MMP-9 ) and the tissue inhibitor of metalloproteinase-1 ( TIMP-1 ) in lung tissues were tested with immunohistochemical analysis. Results 1. The levels of IL-17 in BALF in Pingehuanguben Decoction group and Budesonide group were significantly lower than those in asthma group (all P 〈 0.05 ) , and the IL-17 in BALF was increased compared with the normal con- trol group (P 〈 0. O1 ). 2. The levels MMP-9 ,TIMP-1 and MMP-9/TIMP-1 were decreased in the Pingchuanguben De- coction group and Budesonide group compared with those of asthmatic group ( all P 〈 0.01 ) , and the MMP-9 and MMP-9/TIMP-1 were increased compared with the normal control group( all P 〈 0.01 ). There was no significant diffe- rence between Pingchuanguben Decoction group and Budesonide group ( all P 〉 0.05 ). Conclusions Pingcbuanguben Decoction may restrain airway inflammation and remolding,which can replace Budesonide to a certain extent.
