摘 要:目的:研究白地祛脂合剂对人皮脂腺细胞雄激素受体信使RNA(mRNA)和胞质蛋白表达的影响,探讨中药治疗痤疮的作用机制。方法:采用逆转录-聚合酶链反应(RT-PCR)技术分别检测正常组,低、中、高剂量白地祛脂合剂组作用皮脂腺细胞24h后雄激素受体mRNA的表达情况;采用免疫印迹法(Western Blot)方法检测人皮脂腺细胞胞质蛋白中雄激素受体的表达情况。结果:PCR显示:低、中、高剂量白地祛脂合剂组均使皮脂腺细胞雄激素受体(AR)mRNA表达降低,分别为正常组人皮脂腺细胞的0.592、0.438、0.223倍(P<0.05),呈现一定的量效关系,而高剂量白地祛脂合剂组皮脂腺细胞AR mRNA表达降低最为明显;Western Blot显示:低、中、高剂量白地祛脂合剂组均使皮脂腺细胞雄激素受体胞质蛋白表达降低,分别为正常组人皮脂腺细胞的0.805、0.652、0.550倍(P<0.05),呈现一定的量效关系,而高剂量白地祛脂合剂组皮脂腺细胞AR蛋白降低最为明显。结论:白地祛脂合剂可能通过抑制人皮脂腺细胞AR的表达而具有拮抗雄激素受体对皮脂腺细胞的活性作用,这将为中医药治疗痤疮提供新的治疗途径,有助于从分子层次了解其作用机制。Objective: To study the effect of Baidi Quzhi Mixture on mRNA of human sebaceous gland cells androgen receptor and the expression of cytoplasmic protein, and to explore the mechanism of the treatment of acne by traditional Chinese medicine. Methods: We added different doses of Baidi Quzhi. Mixture in the cultivation of human sebaceous gland cells for 24 hours, and RT-PCR was used to detect the different expressions of androgen receptor mRNA of the normal group, low dose, medium dose and the high dose of Baidi Quzhi Mixture groups. Western Blot was used to detect the expression of androgen receptor in cytoplasmic protein of human sebaceous gland cells. Results: PCR results showed that the low, medium and high doses of Baidi Quzhi Mixture reduced the expression of AR mRNA of the sebaceous cells, it was 0.592, 0.438, 0.223, respectively, and had a dose-effect relationship, the expression of AR mRNA of the high-dose group decreased most significantly (P〈0.05). Western Blot results: the expression of androgen receptor in cytoplasmic protein of human sebaceous gland cells in all groups decreased,and it was 0.805 ~ 0.652, 0.550, respectively, and the high group most significantly. Conclusion: Baidi Quzhi Mixture may have an antagonistic function on the human sebaceous gland cells activation of androgen receptor through the inhibition of expression of human sebaceous cells AR, which will provide a new therapeutic approach to help to understand the molecular mechanisms of Baidi Quzhi Mixture.
