摘 要:目的:观察哮喘豚鼠经β-细辛醚喷雾给药后,对白细胞介素-4(IL-4)、白细胞介素-16(IL-16)和γ-干扰素(γ-IFN)及小鼠免疫器官指数的影响。方法:将豚鼠分为高、中、低剂量组,阳性药组和模型组,复制哮喘模型,造模7d后,喷雾给药,7d后引喘,记录哮喘发作潜伏期和跌倒潜伏期。取血测定IL-4、IL-16和γ-IFN水平。另取小鼠分为高、中、低剂量组,阳性药组和正常组,喷雾给药7d后,摘取胸腺、脾脏,计算免疫器官指数。结果:β-细辛醚能延长哮喘发作潜伏期和跌倒潜伏期(P<0.01);能降低IL-4的含量、提高IL-16和γ-IFN水平,降低IL-4/γ-IFN比值(P<0.05,P<0.01);能提高小鼠胸腺指数和脾脏指数(P<0.05,P<0.01)。结论:β-细辛醚对哮喘有整体治疗作用;有一定的免疫兴奋作用,并有一定的剂量依赖性,其治疗哮喘可能是通过调节Th1/Th2平衡而实现的。Objective: To observe the influence of 13-asarone on interleukin-4 (IL-4), interleukin-16 (IL-16) and interferon-y (y-IFN) level of the asthma model in guinea pig after spray administration, and the impact of the drug on the index of immune organs in guinea pig. Methods: Guinea pigs were randomly divided into five groups: high dose group, medium dose group, low dose group, positive control group and model group to make a model of asthma for 7 days, each group were spray administered for another 7 days and then induced asthma. Asthma attack latency and fall down latency were recorded. Blood samples were collected to measure IL-4, IL-16 and y-IFN level. Furthermore, mice were randomly divided into five groups: high dose group, medium dose group, low dose group, positive control group and normal group. Each group were spray administered. After 7 days, the animals were killed. Thymus gland, spleen were weighed to calculate the index of immune organs. Results: 13-asarone extended the asthma attack latency and fall down latency (P〈0.01), decreased IL-4 level and ratio of IL-4/y-IFN, and increased IL-16 and y-IFN level (P〈0.05, P〈0.01). It also could enhance thymus index and spleen index (P〈0.05, P〈0.01). Conclusion: 13-asarone has overall treatment effect on asthma. The drug has certain immune excited effect. The above effects of fi-asarone has a concentration-dependent manner. The therapy effect of 13-asarone in treating asthma may be regulation of the Thl/ Th2 balance.
